摘要
为胶陀螺菌作为抗氧化剂的开发应用提供参考依据,采用单因素试验与响应面法相结合筛选胶陀螺菌丝体液态发酵最优条件,并在此基础上测定其抗氧化活性。结果表明:最优发酵条件为葡萄糖、酵母浸膏、KH2PO4和MgSO4·7H2O的添加量分别为4.6%、0.3%、0.5%和1%,装液量为100 mL/250 mL,接种量为10%,发酵温度25℃,转速150 r/min摇床培养6 d可获得菌丝体生物量14.95 g/L;菌丝体浓度为1.3 mg/mL时,对DPPH和ABTS自由基的清除率分别达68.25%和78.38%。胶陀螺菌丝体具有较好的抗氧化能力,可作为抗氧化剂产品进行开发。
In order to provide references for the development and application of Bulgaria inquinans as an antioxidant,the optimal conditions of liquid fermentation of B.inquinans were screened by single factor test and response surface method,and the antioxidant activity was determined on this basis.Results:The optimum fermentation conditions were as followed,the adding amounts of glucose,yeast extract,KH2PO4 and MgSO4·7H2O were 4.6%,0.3%,0.5% and 1%,respectively.The liquid volume was 100 mL/250 mL,and the inoculum was 10%,shaken at 25℃,150 r/min conditions for 6 d,and the mycelial biomass would be up to 14.95 g/L.When the mycelium concentration was 1.3 mg/mL,the clearance rates of DPPH and ABTS free radicals were 68.25%and 78.38%,respectively.B.inquinans mycelium has good antioxidant capacity and can be developed as an antioxidant product.
