广西5市伯氏疏螺旋体主要宿主动物鼠和传播媒介的初步调查 被引量:2

Preliminary investigation of rats and other hosts and the vectors of Borrelia burgdorferi in 5 cities in Guangxi Province

摘要 目的调查了解广西伯氏疏螺旋体主要宿主动物鼠和传播媒介感染情况,为有效防控莱姆病提供基础科学数据。方法采用布笼法捕捉野外老鼠并采集其身上寄生的蜱,BSK-II培养基培养从鼠膀胱或蜱中肠中分离到的螺旋体并通过形态学和PCR扩增菌株5s-23s rRNA基因间隔区的方法进行伯氏疏螺旋体鉴定;采用酶联免疫吸附试验(ELISA)检查鼠血清抗伯氏疏螺旋体抗体IgG。结果捕获鼠491只,蜱51只。从鼠体内共分离出16株伯氏疏螺旋体,分离阳性率为3.26%(16/491)。其中斯氏家鼠分离阳性率最高为15.79%(3/19),其次为黄毛鼠,分离阳性率为11.59%(8/69)。莱姆病抗体IgG阳性率为17.92%(88/491),其中斯氏家鼠莱姆病抗体阳性率最高为26.32%(5/19),其次为褐家鼠,阳性率为22.63%(62/274)。蜱体内未检测到伯氏疏螺旋体。结论研究提示在广西5市调查的部分地区存在莱姆病自然疫源地,斯氏家鼠、黄毛鼠、褐家鼠是重要宿主。 Objective To investigate infection with Borrelia burgdorferi in rats or ticks in Guangxi in order to provide basic scientific data to prevent Lyme disease.Method Rats in the wild were caught in cages in 5 cities in Guangxi Province.Parasitic ticks on the rats were collected.B.burgdorferi was isolated from the bladder of rats and the midgut of ticks and subsequently cultured on BSK-II medium.The spirochetes were identified morphologically,and the 5 s-23 s rRNA gene spacer was amplified with PCR.Antibodies against B.burgdorferi in rat serum were examined using ELISA.Result A total of 491 rats and 51 parasitic ticks were collected.Sixteen strains of spirochete were isolated from rats at a rate of 3.26%(16/491).B.burgdorferi was isolated at the highest rate in Rattus sladerni(15.79%,3/19),followed by Rattus losea(11.59%,8/69),Bandicota indica(3.7%,1/27),Rattus flavipectus(3.33%,2/60),and Rattus norvegicus(0.73%,2/274).IgG antibodies against B.burgdorferi spirochetes were detected at a rate of 17.92%(88/491).IgG antibodies were detected at the highest rate in Rattus sladerni(26.31%,5/19),followed by Rattus norvegicus(22.63%,62/274),Rattus losea(20.29%,14/69),Niviventer fulvescens(20.00%,1/5),Bandicota indica(7.41%,2/27),and Rattus flavipectus(6.67%,4/60).B.burgdorferi spirochetes were not found in ticks.Conclusion Results indicated that there are nature epidemic foci of Lyme disease in some districts of the 5 cities in Guangxi province,and its major animal hosts are Rattus sladerni,Rattus losea,and Rattus norvegicus.
