摘要
目的 建立纯度较高的适于试验研究的人绒毛膜滋养层细胞 .方法 胰蛋白酶 - DNase联合消化法培养人绒毛膜滋养层细胞 ,间接免疫染色进行细胞鉴定 ,透射电镜观察细胞内部结构 .结果 倒置显微镜下 ,可见细胞呈不规则多角形 ,核大卵圆形 ,胞质透明 ,呈片状铺展生长 .角蛋白染色阳性率超过 95 % ,未检测到波型蛋白阳性细胞 ,表明不含污染细胞 .透射电镜观察示所获细胞有典型滋养层细胞结构 .结论 该法简便易行 。
AIM To prepare highly purified functional human trophoblast cells from human placentae. METHODS A trypsin DNase dispersion method was used to isolate the trophoblast cells. The cell population was identified by immunocytochemical staining with monoclonal mouse anti human cytokeratin IgG and mouse anti human vimentin IgG. The structural features were revealed by transmission electron microscopy. RESULTS Light microscopic examination revealed patchy spreading and irregular polygonal cells with big and oviform nuclear and transparent cytoplasm. Cytokeratin positive cells were more than 95%. No vimentin positive cells could be detected, indicating absence of contaminating elements. Transmission electron microscopy revealed that the cells had fine structural features typical of trophoblasts. CONCLUSION Trophoblasts may be obtained with the simple method and may offer thereafter a model amenable to experimental manipulation.
