摘要
目的 观察川芎嗪对大剂量甲氨喋呤 (MTX)化疗所致骨髓基质细胞 (BMSC)损伤的保护作用及其量效关系。方法 建立大剂量MTX化疗损伤BMSC的小鼠模型后 ,按每千克体重腹腔注射川芎嗪 0mg(对照组 )、2 0mg(Ⅰ组 )、40mg(Ⅱ组 ) ,1次 /d ,第2 1天采取骨髓细胞进行体外BMSC培养、成纤维细胞集落 (CFU F)计数、BMSC层粘附率测定和BMSC层上造血细胞混合集落培养。结果 BMSC生长、CFU F计数、BMSC层粘附率、BMSC层上造血细胞混合集落数目均表现为Ⅱ组 >Ⅰ组 >对照组 (P <0 0 5 ,P <0 0 1)。结论 川芎嗪对大剂量MTX引起的BMSC损伤有明显的保护作用 。
Objective To evaluate the role of ligustrazine in the damaged murine bone marrow stromal cells (BMSC) after the administration of high dose methotrexate (MTX) and its dose efficacy relationship. Methods After the murine models of BMSC damaged due to high dose MTX were established, each mouse was intraperitoneally injected with ligustrazine of 0 (control group), 20(groupⅠ) or 40 mg/kg(groupⅡ) once a day. On the 21st day bone marrow cells were harvested, then BMSCs were cultured; number of colony forming unit fibroblast (CFU F) was counted; the adhesive rate of BMSC was assayed and the growth of colony forming unit mixed hematopoietic cells (CFU Mix) on the BMSC layer was observed in vitro . Results The number of CFU F and the adhesive rate of BMSC layer in group Ⅰ or group Ⅱ were significantly higher than those in control group ( P <0.05 or P <0.01), and those in group Ⅱwere also significantly higher than those in group Ⅰ( P <0.05 or P <0.01). The growth of BMSC or CFU Mix on the BMSC layer in group Ⅱ was faster than that in groupⅠ, and that in groupⅠ grew faster than that in control group. Conclusion Ligustrazine can significantly protec the damaged BMSC caused by high dose MTX in dependence on its dose.
