摘要
目的 :探讨 p2 7kip1 、PCNA及 p1 6与胃肠道类癌转移的关系。 方法 :采用免疫组化SABC法检测 2 5例胃肠道类癌组织中p2 7kip1 和PCNA蛋白表达 ,同时用PCR法检测 p1 6基因的缺失情况。 结果 :p2 7kip1 在胃肠道类癌阳性表达率为 64 % (1 6/ 2 5) ,PCNAⅠ~Ⅱ级和Ⅲ~Ⅳ级表达率分别为 56 %、44 %。 2种蛋白阳性表达率与分化程度无相关性 (P >0 0 5) ,与淋巴结转移相关 (P <0 0 5)。p2 7kip1 与PCNA表达负相关。p1 6基因纯合性缺失率为 48% (1 2 / 2 5) ,其缺失率与类癌分化程度和淋巴结转移无关。结论 :p2 7kip1 低表达、PCNA高表达与胃肠道类癌转移有关 ,胃肠道 ;类癌的发生与p1 6基因缺失有关 ,但可能为早期分子事件。
Purpose To study the significance of p27 kip1 ,PCNA protein expression and loss of p16 gene in gastrointestinal carcinoid tumors. Methods The expression of p27 kip1 ,PCNA was detected by SABC immunohistochemical technique in 25 cases of gastrointestinal carcinoid. Loss of p16 gene was detected by PCR method. Results The positive rate of p27 kip1 protein was 64%,and PCNA expression rates at grade Ⅰ Ⅱ, and grade Ⅲ Ⅳ were 56%,44%,respectively. The expressions of p27 kip1 and PCNA were correlated with lymph node metastatsis. Loss of p16 gene was found in 12 cases (48%), which was not associated with tumor differentintion and lymphode metastasis. Conclusion The down regulation of p27 kip1 expression and high expression rate of PCNA are correlated with lymphnode metastasis, and may be used as prognostic indicators in gastrointestinal carcinoid tumors. Loss of p16 gene is correlated with pathogenesis of gastrointestinal carcinoid tumors, and may be a early event.
