Plant Regeneration from Corn Protoplast Culture

摘要 将玉米自交系“330”成熟胚诱导的愈伤组织,在附加2mg/L2,4—D的MSB培养基上继代、筛选,产生了三种类型的愈伤组织。用结构疏松,生长迅速的Ⅱ型胚性愈伤组织分离原生质体,在KPR或N6ap培养基中以1×105/ml密度进行液体浅层培养,植板率为5.6%—7.4%。采用分步分化法得到了正常的绿苗,并经过壮根,炼苗后移栽成活。 Three types of calli were derived from the mature embryo cultures of an important commercial maize inbred line '330', which were cultured on MSB medium with 2mg/L2,4—D. Protoplasts were digested from embryogenic calli of type Ⅱ that were friable, no—nucilaginous and repidly growing. Plating frequency of 5.6%—7.4% was obtained when protoplasts were cultured in KPR or Neap medium by thin liquid layer culture method at density of 1×10~5 cells/ml. Plant regeneration was achieved with step by step differentiation method. After being transplanted into soil, plants were recovered and grown normally.
