^(60)Co γ 线照射大鼠肥大细胞的定量分析

Quantitative analysis of mast cell in 60 Co γ-ray irradiated rats

摘要 用新建立的阿里新蓝-番红花红染色结合图像分析,观察了正常和60Coγ线照射大鼠肥大细胞(MC)数量和定量参数的改变,发现:(1)正常大鼠皮肤组织MC数量最多,胞体较大,胞浆颗粒密度和数量均高于肝脏;胃组织MC数量最少,胞体介于皮肤和肝脏之间,然而其颗粒密度最高,颗粒数量仅次于皮肤;肝脏MC数量居中,胞体较小,其颗粒密度和数量均低于皮肤和胃组织。(2)照射后各组织MC数量出现一致增加,其颗粒密度和数量也出现规律性变化:如照后3和9个月,皮肤和肝脏MC颗粒密度和数量均明显高于对照组,提示此时期激活的MC具有旺盛的合成和释放颗粒的能力;而在照后6和9个月,颗粒密度和数量均出现降低,推测此时期可能为颗粒合成低峰期。胃组织在照后3-9个月,其颗粒密度和数量均持续降低,表明胃组织MC释放颗粒的形式及合成颗粒的能力均不同于皮肤和肝脏。上述定量分析数据对于进一步探讨MC辐射损伤特点及其参与放射性器官纤维化形成的机理均具有重要意义。 The numberal changes and morphometric parameters of mast cells (MS) in normal and 60 Co γ-ray irradiated rats were observed by using histological method of alcian blue-safranin combined with image analysis. The results showed that: (1) In normal rats, the number of MC and their cytoplasmic granules were much more, and the area of MC and their cytoplamic granular density were larger in skin than those of the liver. The number of MC in stomach was the least, the area of MC was in moderate degree, however the MC granular density was the highest among three tissues. The number of MC in live was in the middle, the MC area was smallest, and the density and quantity of cytoplasmic granules were lower than those of the skin and stomach. (2) After irradiation, the total MC in all tissues increased identically and the cytoplasmic granular density and quantity changed regularly. For instance, the density and number of MC cytoplasmic granules in skin and live were higher evidently than those of control group on the third and nineth months after irradiation, suggesting that MC activated with potential ability to synthesize and release granules during these period of the time; while on the sixth and nineth months after irradiation, the cytoplasmic granular density and quantity decreased obviously, conjecturing that the ability to synthesize granules was lower in this time. However the density and number of the gastric MC granules decreased gradually, indicating that the mode of realeasing and the ability of synthesizing granules of gastric MC were different from those of skin and liver. The results mentioned above are consistent with the formation of radiation-induced organic fibrosis in live, and suggest that MC and their granular mediated effect may plays an important role in the formation of organic fibrosis.
