摘要
目的 对藁本药材的化学及药效学进行研究 ,确定藁本药材质量评价的指标并建立评价方法。方法 采用柱色谱的方法 ,从中国藁本及辽藁本的根及根茎中共分离得到 17个化合物。根据藁本药材的功能主治设计了抗炎、镇痛、镇静等药效学实验 ,对藁本药材粗提物及单一化合物进行了实验 ,并将实验结果进行了比较 ,确定了能够代表藁本药材功能主治的指标成分 ,建立了该指标成分的HPLC定量分析方法 ,分析了 2 0个产地的藁本药材。结果 藁本药材中主要成分阿魏酸是能够代表藁本药材功能主治的指标成分 ,而且在 2 0个产地中均含有 ,可以做为评价藁本药材质量的指标成分。结论 中草药的质量控制问题是中草药现代化研究的瓶颈问题 ,本实验为此方面的研究提供了可借鉴的方法。
OBJECTIVE: To study the chemistry and pharmacology of Rhizoma and Root Ligusticum, and to find out the constituents for quality control as well as to establish the analytical method for its quality control. METHOD: Seventeen constituents were separated and identified from medicinal herb Ligusticum by column chromatography. According to the clinical efficacy of genus Ligusticum, the pharmacological models of anti-infection, sedation and analgesia were established. The extracts of rhizoma Ligusticum and constituents were tested on these models and the results were compared. The indicator for the clinical efficacy of genus Ligusticum was found out and the HPLC method for its determination was established. Twenty samples from different habitats were analyzed. RESULT: Ferulic acid was the constituent whose pharmacological activity represented the clinical efficacy of genus Ligusticum and was found in twenty samples. It appeared to be suitable as the constituent for quality control of genus Ligusticum. CONCLUSION: The quality control of Chinese Herb Medicine is the key problem in study of modernization of Chinese Herb Medicine. The paper provides the feasible method and idea for the modenization of Chinese Herb Medicine.
