摘要
目的 研究小鼠微小残留白血病模型的建立方法。方法 采用P388白血病细胞系和DBA小鼠建立P388/DBA小鼠微小残留白血病模型。结果 P388细胞腹腔接种DBA小鼠 ,白血病的发病率为 10 0 % ,无自发缓解。P388白血病模型对环磷酰胺 (Cy)敏感 ,有较好的药物剂量 生存时间关系 ,接种细胞数量与Cy剂量间有良好回归关系 ,是研究微小残留白血病的理想模型。移植生物试验提示 ,当微小残留白血病细胞数量在 10 1左右时 ,其分布可能局限于肝、脾及股骨骨髓以外的器官和组织中。
Purpose. To study establishing a model of mininmal residual leukemia in mice. Methods. P388/DBA model of minimal residual leukemia was established by using P388 leukemic cell line and DBA mice. Results. P388 cells were inoculated in the abdomenal cavities of DBA mice, the incidence of leukemia was 100 percent, Any mice with leukemia could not obtain remission spontaneously. The model of leukemia was sensitive to cyclophosphamide (Cy) and the time of survival was related to the dose of Cy received. The number of cells inoculated in mice and the dose of Cy were correlated regressionally. So this model was ideal to be used to do some research on minimal residual leukemia. The results of biological transplantation experiments indicated that the leukemic cells probably existed in organs and tissues except liver, spleen and femur bone marrow of mice, when the number of residual leukemic cells was no more than 101. Conclusions. P388/DBA model of minimal residual leukemia could be established by using P388 leukemic cell line and DBA mice.
