人体失神经支配骨骼肌Na,K-ATP酶、Ca-ATP酶及糖原的变化 被引量:2

摘要 目的 探讨失神经支配人体骨骼肌酶和糖原改变。方法  16例不同时间人体失神经骨骼肌与 3例正常骨骼肌标本 ,用组织化学方法测定Na,K ATP酶和Ca ATP酶含量 ,电镜和PAS染色观察糖原分布改变并测定糖原含量。结果 人体骨骼肌随失神经支配时间延长 ,Na ,K ATP酶和糖原含量呈进行性下降趋势 ;而Ca ATP酶在失神经 2个月内下降约 1/ 3,3个月后一直维持在正常值的 30 %~ 4 0 %。PAS染色显示人体骨骼肌失神经后出现PAS染色阴性细胞 ,其数量在 3~ 6个月时达到高峰 ,随后逐渐下降 ;在PAS染色切片中 ,居中肌细胞核主要在阴性肌纤维中出现。结论 Ca ATP酶活性变化规律说明失神经支配后肌肉收缩的生理基础长期存在 。 Purpose To study the change of enzyme activity and glycogen of denervated human skeletal muscle. Methods 16 cases denervated and 3 cases normal human skeletal muscles were involved in this experiment.The activities of Na,K?ATPase,Ca ATPase and were measured histochemically.The distribution of glycogen granules was observed by PAS staining and electron microscopy. Results Activities of Na,K?ATPase,Ca ATPase, and the contents of glycogen declined progressively along with the post denervated time.But Ca ATPase activity reduced 1/3 of normal value at 2 months,then remained 30%-40% of its activity.There were PAS negative myofibers in the denervated human s ke l etal muscle,its quantities reached to a high peak at 3-6 months and then reduced along with the post denervated time.The central myonuclei could be found in the PAS negative m yofibers. Conclusions Physiologic basis of muscle constraction may exist for a long period after its denervation.The PAS negative myofiber may be a reliable index of regeneratin of denervated human skeletal muscle.
