不同诱变处理对紫花苜蓿生理生化指标的影响 被引量:4

摘要 为了比较研究不同诱变处理方式对紫花苜蓿诱变效果的影响。试验通过^(60)Co-γ射线、紫外线、甲基磺酸乙酯(EMS) 3种诱变处理对供试4种紫花苜蓿干种子进行诱变,以当代材料为研究对象,探讨诱变处理对苜蓿生理生化指标的影响。诱变处理后,3种抗氧化酶活性以增加趋势为主。其中对SOD的影响,^(60)Co-γ表现为促进作用,紫外线处理降低了SOD活性,EMS处理表现为低促髙抑作用;3种诱变处理对POD和CAT的影响为^(60)Co-γ辐射随处理剂量增加酶活性下降,紫外线处理则随着剂量增加酶活性增加,EMS处理则因品种不同而不同。诱变处理对苜蓿可溶性蛋白、可溶性糖和游离脯氨酸含量的影响随剂量及品种变化而不同,对MDA含量影响不显著。不同诱变处理显著提高了苜蓿叶片抗氧化酶活性,其中150 Gy^(60)Co-γ、90 min紫外线和0.4%EMS处理对苜蓿生理活性具有明显提高作用。 This study aims at comparing the effects of different mutagenesis treatments on alfalfa.^(60) Co-γ rays,ultraviolet and EMS were used for induced mutation of four alfalfa cultivars' dry seeds. The contemporarymaterials were taken as research objects, the influence of mutagenic treatment on the physiological andbiochemical indexes of alfalfa was explored. After mutation treatments, the activity of three anti-oxidativeenzymes showed an increasing trend. The effect of^(60) Co-γ irradiation on SOD was accelerating, ultraviolettreatment reduced SOD activity, EMS treatment on SOD showed low-concentration promotion and high-concentration inhibitory. Along with the increasing of^(60) Co-γ irradiation, POD and CAT activity decreased,while POD and CAT activity increased with the increasing ultraviolet dosage. The effect of EMS treatment wasdifferent in different varieties. The effects of mutagenic treatments on soluble protein, soluble sugar and freeproline in alfalfa were different due to dosage and variety, and did not affect MDA content obviously. Themutation treatments improved the activities of three anti-oxidative enzymes, 150 Gy^(60) Co-γ, 90 min ultravioletand 0.4% EMS treatment had significant effect on the physiological activity of alfalfa.
