子痫前期与GNA12启动子甲基化的相关性分析 被引量:3

摘要 目的探讨鸟嘌呤核苷酸结合蛋白(GNA12)启动子甲基化程度与子痫前期(pre-eclampsia,PE)的关系。方法选取2016年8月至2017年7月宝鸡市妇幼保健院收治的PE患者共50例作为研究组,另取同期行正常孕检的孕妇50例作为对照组,通过Massar Ray核酸质谱分析系统分析两组胎盘外周血DNA样本中GNA12启动子8个Cp G位点的甲基化水平及m RNA表达情况。结果研究组胎盘DNA样品中,73位点、109位点、185位点及204位点的GNA12甲基化程度显著小于对照组(P<0.05),外周血样品中73位点和185位点的GNA12甲基化程度显著小于对照组(P<0.05)。研究组GNA12基因的m RNA表达水平显著高于对照组(P<0.05)。结论 PE与GNA12启动子的甲基化水平降低相关,可对胎盘和外周血进行基因检测,且外周血检测更加便捷,对PE的早期诊断有一定的临床意义。 Objective The guanine nucleotide binding protein(GNA12) is overexpressed in pre-eclampsia, this study was designed to investigate the potential association of GNA12 promoter methylation with pre-eclampsia. Method A total of 50 patients with PE were enrolled in Baoji Maternal and Child Health Hospital from August 2016 to July 2017 as the study group. 50 pregnant women with normal pregnancy test were selected as the control group. The methylation level and mRNA expression of the eight CpG sites of the GNA12 promoter in the two groups of placenta and peripheral blood DNA samples were analyzed by Massar Ray. Result The study group placental DNA samples, the degree of methylation of 73 GNA12 site, 109 site, 185 site, 204 site was significantly less than the control group(P〈0.05). Study peripheral blood samples, 73 and 185 degree points GNA12 methylation point significantly less than the control group(P〈0.05). The m RNA expression level of GNA12 gene in study group was significantly higher than that in control group(P〈0.05). Conclusion Methylation level GNA12 pre-eclampsia and related reduction promoter, placenta and peripheral blood may be obtained by genetic testing, has clinical significance for the early diagnosis of PE.
