补益营卫方对衰老表皮脂质代谢的影响 被引量:3

摘要 目的:研究补益营卫方对衰老表皮脂质代谢的影响,探讨其延缓皮肤衰老,改善表皮脂质稳态的可能机制。方法:消化分离出年轻小鼠与自然衰老小鼠表皮细胞并传代培养,其中衰老小鼠表皮细胞分为两组,一组常规培养,一组用含2.5%补益营卫方培养基培养。荧光定量RT-PCR检测各组细胞HMGCR、LASS4、APOE和APOD的mRNA表达。结果:与年轻组比较,老年组表皮细胞中HMGCR、LASS4、APOE和APOD mRNA表达水平显著升高(P〈0.05);经药物干预后,衰老表皮细胞中HMGCR、LASS4、APOE和APOD mRNA表达水平较老年组显著降低(P〈0.05)。结论:补益营卫方可能通过对衰老表皮脂质代谢相关基因HMGCR、LASS4、APOE和APOD mRNA等的影响,调节衰老表皮脂质代谢,起到延缓皮肤衰老、改善表皮脂质稳态的作用。 Objective: To study the effect of Recipe of Supplementing Ying-wei on the lipid metabolism of senescent epidermis. The mechanism of the improvement of epidermal lipid homeostasis is discussed. Methods: We separately cultured epidermal cells from young and natural aging mice then subcultured,including the epidermal cells of aging mice were divided into two groups. One group was conventional cultivation and the other group was cultured in a medium containing 2. 5 percent Recipe of Supplementing Ying-wei. The mRNA expression of HMGCR,LASS4,APOE and APOD were detected by fluorescent quantitative RT-PCR in each group. Result: Compared with the younger group,the expression levels of HMGCR,LASS4,APOE and APOD mRNA in the epidermal cells of elderly group were significantly increased( P〈 0. 05). The expression levels of the epidermal HMGCR,LASS4,APOE and APOD mRNA were significantly reduced in senescent epidermal cells( P〈 0. 05),compared with those of the older group + 2. 5% drug group. Conclusion: The effect of HMGCR,LASS4,APOE and APOD mRNA may be used to regulate the lipid metabolism of aging epidermis by Recipe of Supplementing Ying-wei which can delay the aging of skin and improve epidermal lipid homeostasis.
