丹皮酚对裸鼠肝癌原位移植瘤凋亡、NF-κB p65及Caspase-9的影响 被引量:1

摘要 目的:研究丹皮酚对肝癌原位移植瘤组织凋亡及半胱氨酸天冬氨酸蛋白酶-9(Caspase-9)、核转录因子NF-κB p65的影响。方法:用MHCC-97H人肝癌细胞建立裸鼠肝癌原位移植瘤,之后将50只造模成功裸鼠随机分入生理盐水组、5-氟尿嘧啶组、丹皮酚低剂量组、中剂量组、高剂量组,分别腹腔注射生理盐水、5-氟尿嘧啶、丹皮酚20 d后,取瘤体组织。免疫组化法原位检测组织细胞凋亡及Caspase-9、NF-κB p65的表达,并结合Image-Pro Plus 6.0图像处理系统分析免疫组化结果。结果:与生理盐水组相比,丹皮酚低、中、高剂量组及氟尿嘧啶组凋亡程度均有不同程度升高,差异有统计学意义(P〈0.05),丹皮酚高剂量组凋亡最显著。丹皮酚低、中、高剂量组和氟尿嘧啶组Caspase-9阳性表达强于生理盐水组,NF-κB p65表达弱于生理盐水组。丹皮酚中剂量组Caspase-9、NF-κB p65的表达与氟尿嘧啶组无明显差异(P〉0.05)。结论:丹皮酚可以诱导裸鼠体内肝癌细胞凋亡,其作用机制可能是通过上调Caspase-9的表达,同时抑制NF-κB p65而实现。 Objective:To investigate the effect of paeonal on apoptosis and the expression of NF-κB p65,Caspase-9 transplanted tumor of human hepatocellular carcinoma in nude mice. Methods:We apply MHCC-97 human liver cancer cells to establish nude mouse orthotopic transplantation of liver cancer tumor,since then 50 building successful nude mice were randomly drawn in blank model group,the 5-fluorouracil group,paeonol low dose group,middle dose group,high dose group,and were intraperitoneal injected with Paeonal,5-fluorouracil,for 20 days. The apoptosis and the expression of NF-κB p65,Caspase-9 were tested by immunohistochemistry,connecting with the Image-Pro Plus 6.0. Results:Compared with normal saline group,the apoptotic degree of paeonol low,medium and high dose group and fluorouracil group were significantly increased(P〈0.05),the apoptotic expressed strongest. The positive expression of Caspase-9 in paeonal low,medium and high dose group and fluorouracil group were stronger than that in saline group,and NF-κB p65 expression was weaker than that of saline group. There was no significant difference in the expression of Caspase-9 and NF-κB p65 between the paeonol low dose groups and the fluorouracil group(P〈0.05). Conclusion:Paeonal can induce liver cancer cell apoptosis in nude mice and its mechanism may be through the increase the expression of Caspase-9 and inhibit the expression of NF-κB p65.
