补阳还五汤对急性脊髓损伤大鼠内质网应激相关因子影响的研究 被引量:15

Effect of Buyanghuanwu Decoction on the Expression of Endoplasmic Reticulum Related Factors after Acute Spinal Cord Injury in Rats

摘要 目的:探讨补阳还五汤灌胃治疗对大鼠急性脊髓损伤(Spinal Cord Injury,SCI)的修复作用及作用机制。方法:将SPF级3月龄Wistar雌性大鼠40只,随机分成假手术组、模型组、甲强龙组、补阳还五汤组(中药组)。造模后第1天,甲强龙组经尾静脉注射甲强龙治疗共24h,首剂量为30mg/kg,余以5.4mg/(kg·h)每4h给药1次;中药组以中剂量补阳还五汤颗粒剂,配制成含颗粒剂2.00g/mL的水溶液,每天3.5g/kg灌胃,相当于成人用量的1倍,1次/d,共14d.治疗后第1,3,7及14天分别以BBB分级法评价神经功能恢复情况,Western Blot法检测SCI节段Caspase-12,Caspase-9及Caspase-3蛋白的表达。结果:各时间点BBB评分随时间延长而上升,差异有统计学意义(P<0.01);治疗后7d和14dBBB评分比较,差异无统计学意义(P>0.05)。组间比较:中药组和甲强龙组的BBB评分高于模型组,差异有统计学意义(P<0.01);中药组和甲强龙组比较差异无统计学意义(P>0.05)。治疗后第14天大鼠脊髓组织中Caspase-12,Caspase-9及Caspase-3蛋白的表达:中药组和甲强龙组各蛋白表达均低于模型组,差异有统计学意义(P<0.01);与假手术组比较,差异无统计学意义(P>0.05);中药组和甲强龙组比较差异无统计学意义(P>0.05)。结论:围手术期以补阳还五汤治疗急性SCI,对大鼠脊髓损伤具有修复作用,其作用机制可能是通过抑制过度的内质网应激反应,下调Caspase-12,Caspase-9及Caspase-3蛋白的表达。 Objective:To investigate the repair effect and mechanism of Buyanghuanwu decoction(BYHWD)on spinal cord injury(SCI)in rats.Methods:Forty female SPF Wistar rats were randomly divided into sham operation group(SO),model group,methylprednisolone group(MP),and BYHWD group,with 10 rats in each group.On the first day after model establishment,the MP group was treated with methylprednisolone injection through the tail vein for a total of 24 h.The first dose was 30 mg/kg,and then was administered with 5.4 mg/(kg·h)once,interval by every 4 h.The rats in BYHWD group were given middle dose of BYHWD at a final concentration of 2 g/mL,with BYHWD administration at 3.5 g/kg once a day(09:00)for 14 d,which was equivalent to 1 times adult dosage.Nerve functional recovery was evaluated by BBB score method at 1 st,3 rd,7 th and 14 th d after treatment,and the protein expression of Caspase-12,Caspase-9 and Caspase-3 in SCI tissue was detected by Western Blot method.Results:The results of BBB score showed that BBB score were increased as time increased(P〈0.01).There was no statistically significant difference between 7 dand 14 d(P〉0.05).The BBB score in BYHWD group and MP group were higher than that in model group(P〈0.01).There are no significant difference between BYHWD group and MP group(P〉0.05).The results of protein expression showed that the expression of Caspase-12,Caspase-9,and Caspase-3 in BYHWD group and MP group was significantly lower than that in the model group(P〈0.01),which was no significant different with sham group(P〉0.05).There was no significant difference between BYHWD group and MP group(P〉0.05).Conclusion:BYHWD is effective for treating acute SCI during preoperative time point.Its mechanism may be through the inhibition of excessive endoplasmic reticulum stress reaction and down regulation of Caspase-12,Caspase-9 and Caspase-3 protein.
