摘要
目的 用脂多糖 (LPS)诱导性实验牙周炎模型观察大黄素抑制LPS对动物牙周组织的毒性效应。方法 建立LPS诱导性鼠牙周炎模型 ,18只大鼠随机分为T组 (实验组 )和C组 (对照组 )各 9只 ,T组牙周炎部位局部注射 0 1mg/ml大黄素溶液 0 4ml,C组局部注射等量生理盐水 ,均连续 3天。分别在用药后 0、3、7、14天处死取样 ,用光镜、透射电镜观察。结果 用药后 3、7、14天 ,C组牙周组织炎症反应明显重于T组。透射电镜下 ,T组用药后 7天牙龈成纤维细胞内的线粒体轻度肿胀 ,粗面内质网扩张 ,胶原排列正常 ,而C组牙龈成纤维细胞的线粒体肿胀、基质空化、严重者线粒体膜破裂 ,粗面内质网扩张、囊泡化 ,胶原崩解。结论 大黄素能阻断LPS对动物牙周组织损害 。
Objective To study the inhibitory effect of emodin on the toxicity of bacterial endotoxin(LPS) in rats models of periodontal disease.Methods In this protocol,we used 18 adult male SD rats with experimental periodontitis induced by repeated injection of LPS into the maxillary labial and palatal gingiva between first and second upper molars for 3 days in succession.After 1 week,these rats were divided randomly into 2 groups:T and C.Each contained 9 rats and one of them was used as a blank control.We respectively injected 0 1mg/ml emodin solution into the rats periodontal destructive tissue of T group and the physiological saline into these of C group and repeated infection for 3 days.The rats were sacrificed at different periods (0,3,7,14d) and periodontal tissues were observed by histology.Results After 3,7,14d of the test,the ratsperiodontal inflammation of T group was significantly slighter than that in C group.Ultrastructurally,the changes of fibroblasts in C group included hydropic mitochondria,obvious cavitation of the mitochondria matrix and loss of cristae and mitochondria breakage;dilation and vesiculation as well as degranulation of rough endoplasmic reticulum.But the changes of fibroblast in T group mentioned above were relatively slight.Conclusions The study suggests that emodin can inhibit destructive effect of endotoxin to the rats periodontal tissue.
