摘要
采用乙醇溶剂法提取大黄鱼鱼卵(籽)磷脂的工艺条件。通过单因素试验研究乙醇体积分数、浸提次数、料液比、浸提时间和浸提温度对磷脂提取率的影响。在此基础上,采用Box-Behnken设计方法进行响应曲面法试验设计,确定最佳工艺:乙醇体积分数95%,浸提3次,料液比1∶10 g/m L,浸提时间30 min,浸提温度40℃。在此条件下,鱼卵粉磷脂提取率的平均值为(62±0.38)%,接近模型预测值。采用气相色谱法分析大黄鱼鱼卵(籽)磷脂的脂肪酸组成,结果表明,磷脂中含有20种脂肪酸,多不饱和脂肪酸有9种,占总脂肪酸含量的17.35%,其中EPA和DHA占总脂肪酸含量的15.19%。
Ethanol solvent was used to extract Phospholipids from Large yellow croaker fish egg. Extraction times, ethanol concentration, liquid-to-solid ratio, ethanol concentration and extraction temperature were researched by a series of single-factor experiments. Based on single factor test, response surface design was used through the Box-Behnken De- sign principle, the best extraction conditions are as follows: ethanol concentration was 95%, extraction times were 3, liquid-to-solid ratio was 1:10, extraction time was 30 min, and extraction temperature was 40 ~C. Under these condi- tions,the product yield of phospholipids was (62 + 0.38)%, and this result is closed to predicted value. The fatty acids of phospholipids were analyzed by GC, the results show that the fatty acids were composed by 20 kinds of fatty acid, including 9 kinds of polyunsaturated fatty acids, accounting for 17.35% to the total fatty acid. EPA and DHA accounted for 15.19% to the total fatty acids.
