摘要
目的 :检测大鼠肝脏损伤后 3种神经营养因子的mRNA表达的规律 ,为进一步探讨肝脏再生的分子机制提供新线索。方法 :提取成年大鼠部分肝切除后再生肝中的总RNA ,利用半定量RT -PCR法分析BDNF ,GDNF和FGF - 2基因在不同时间点的表达水平和变化规律。结果 :GDNFmRNA在正常时几乎检测不到 ,部分肝切除后 ,其表达水平明显上升。BDNF和FGF - 2的mRNA水平在手术后也同样有显著的升高。结论 :以上结果表明 ,这三个神经营养因子可能参与了肝再生的过程。
Objective:To investigate the molecular mechanisms of liver regeneration and to search new clues for improving liver regeneration following injury. Methods:The alteration of mRNA expression levels of three trophic factors in liver following liver excision was studied using highly sensitive semi-quantitative RT-PCR.Results:The expression levels of two spliced forms of GDNF gene were increased following injury. Marked increases of BDNF and FGF-2 were also observed in the regenerating liver 24~72 hr after liver excision.Conclusion:These findings suggest that BDNF, GDNF and FGF-2,which are known to be associated with neural growth and development, may be involved in liver regeneration.
