摘要
目的 :观察脑出血模型大鼠脑内脑源性神经营养因子 (BDNF)的分布以及中药脑溢安对其影响。方法 :通过微量注射器向苍白球内注入Ⅶ型胶原酶 0 .4U建立脑出血模型 ,用免疫组织化学法观察脑出血后 2h ,6h ,12h ,1d ,4d ,7d ,6个时间点BDNF的表达变化 ,以阳性细胞计数作为观察指标。结果 :脑出血后 2h ,BDNF主要表达于血肿周围的小胶质细胞 ,6h表达开始增高 ,12h后达高峰 ,以后逐渐降低 ,到第 7天模型组和脑溢安组仍有表达 ,其中在 12h ,1d ,4d ,3个时间点上 ,两组阳性细胞计数比较差异有显著性 (P <0 .0 5 )。结论
Objective To observe the distribution of brain derived neurotrophic factor (BDNF) protein in rat brain with cerebral hemorrhage and effect of Naoyi an on BDNF. Methods The intracerebral hemorrhage (ICH) animal models were established by microinjecting 0.4 U Ⅶ collagenase into the globus pallidus. We observed the expression variation of BDNF at 2 h, 6 h, 12 h, 1 d, 4 d and 7 d after cerebral hemorrhage by immunohistochemistry, and positive cell counting served as the observation index.Results BDNF mainly expressed in microglia around the perihematom after ICH, BDNF began to express at 2 h after cerebral hemorrhage, and rose at 6 h, and peaked at 12 h. At the 7th day, BDNF still expressed in both groups. At 12 h, 1 d, and 4 d, positive cell counting of the two groups had significant difference (P<0.05).Conclusion Naoyi an can promote the expression of BDNF protein after ICH.
