摘要
2-苯乙醇(2-phenylethanol,PEA)是一种具有玫瑰香气的芳香醇,广泛应用于食品、化妆品和药品等领域。2-苯乙醇对酵母的抑制作用是生物法合成2-苯乙醇的主要瓶颈,本实验通过紫外诱变技术对酿酒酵母AS2.516进行诱变育种,筛选获得了一株2-苯乙醇耐受性和产量均有明显提高的菌株XZ-33,转化合成2-苯乙醇浓度达到3.55 g/L,比原始菌株提高了19.5%,摩尔转化率达到95.7%。
2-phenylethanol(2-PEA) as an aromatic alcohol with a rose-like fragrance was widely used in food,cosmetic and pharmaceutical industry.The inhibit ion of 2-phenylethanol to yeast is the main difficult problem in the production of 2-phenylethanol by Saccharomyces cerevisiae.In this study,Saccharomyces cerevisiae AS2.516 was treated by Ultraviolet radiation.As a result,a mutated strain designated XZ-33 was screened out,both tolerance and yield increased significantly,gained the yield of 2-phenylethanol at 3.55 g/L,increased by 19.5% compared with the original strain,the molar conversion rate of L-phenylalanine to 2-phenylethanol is 95.7 %.
