摘要
为研究牛包虫病基因工程亚单位疫苗EG95对牦牛的免疫效果、安全性,并调查该疫苗在不同性别、不同年龄、不同海拔地区牦牛间存在的免疫差异性。对4月龄及以上牦牛以免疫剂量每次1头份,每头份含EG95 250μg进行首免、加免(28d后)、安全评价(免疫剂量每次2头份含EG95 500μg),在试验前、首免后28d及加免后28d分别对免疫效果组和对照组牦牛采血,应用间接ELISA法检测血清中特异性抗体。结果显示,首免后28d免疫抗体群体阳性率为71.33%,加免后28d免疫抗体群体阳性率为83.00%,相比首免免疫抗体群体阳性率提高11.67%,与对照组、免疫前比较差异显著(P<0.05)。安全试验组牦牛免疫后第1天有6头出现精神沉郁、采食量下降、体温升高、注射部局部肿胀等不同程度免疫应激反应,4d后均恢复正常。石渠县、康定市不同海拔地区试验牦牛在免疫抗体产生时间、群体阳性率等方面,无明显差异(P>0.05)。结果表明,该疫苗适用于不同海拔地区牦牛使用,安全并可产生较高的抗体水平,对4月龄及以上牦牛免疫程序基本为2次免疫,分别颈部皮下注射1头份,间隔期28d。
To study immune effect and securityof the genetic engineering subunit vaccine EG95 of bovine echinococcosis,and to investigate immune difference of sex, age, and altitude yak. for more than 4 months of age and yak immune dose every time 1 head,a head of containing EG95 (inclu ding 250 g for the first free,free (after 28 d),safety assessment (immune dose every 2 head of EG95 (including 500 g) ,before the test,after the first free 28 d and 28 d after add free group and the control group on the immune effect yak blood respectively, application of indirect ELISA method todetect specific antibody in serum. 28 d after the first free immune antibody positive rate was 71. 33% ,and 28 d after free immune antibody positive rate was 83.00% ,compared with the first free immune antibody group were increased by 11.67%,compared with control group,the immune be- fore significant difference (P〈0.05). Yak immunization safety test group 1 d after six depressed head appear spirit, decreased feed intake, body temperature, injection of local swelling degree of dif- ferent immune stress reaction,are back to normal after 4 d. Yak Shi Qu county,kangding city dif- ferent altitudes test in such aspects as immune antibody generation time, group were, no significant difference (P〉0.05). The vaccine is suitable for different altitude yak use,safe and can produce a higher level of antibodies, for more than 4 months of age and yak immune program basic for 2 times, respectively, 1 head, neck subcutaneous injection interval 28 d.
