基于脑区特异性不同剂量冰片与川芎配伍抗脑缺血的研究 被引量:12

Research on the synergy of different doses of borneol and Chuanxiong Rhizoma on the anti-ischemnia effect in different brain regions

摘要 目的:观察不同剂量冰片配伍川芎对不同脑区缺血的保护作用。方法:复制大鼠全脑缺血-再灌注模型,观察不同剂量冰片(0.01,0.02,0.04,0.08,0.16,0.32,0.64,1.28 g·kg-1)联合川芎对大鼠皮层、海马、下丘脑和纹状体四个脑区梗死率,脑组织IL-1β、IL-6、TNF-α含量和组织学评分的影响。结果:川芎单用能显著改善皮层区TNF-α含量。其配伍冰片后对四个脑区的梗死率,IL-1β、IL-6、TNF-α含量和组织学评分均表现出更好的疗效。另外针对不同脑区,两药配伍疗效比较为皮层>海马、纹状体>下丘脑。针对冰片剂量,两药配伍疗效比较为0.08 g·kg^(-1)>0.16 g·kg^(-1)>0.32 g·kg^(-1)。结论:川芎和冰片配伍可以对缺血脑组织产生脑区特异性改善,而且这一改善作用与冰片剂量密切相关。 OBJECTIVE To investigate the enhancement effects of borneol at different doses on the anti-ischemnia of Chuanxiong Rhizoma(CR)in different brain regions.METHODS After the model of cerebral ischemia-reperfusion was established,the effect of CR combined with different doses of borneol was evaluated based on brain infarct,levels of IL-1β,IL-6,TNF-αand nissl staining.RESULTS Monotherapy of CR only improved TNF-α content in cortex,but the combined therapy of CR and borneol produced better improvements in infarct area,levels of IL-1β,IL-6,TNF-α and nissl staining in each brain region.Moreover,the therapeutic effects according to the brain regions were cortex〉hippocampus,striatum〉hypothalamus,the therapeutic effects according to the borneol dose were 0.08 g·kg^-1〉0.16 g·kg^-1〉0.32 g·kg^-1.CONCLUSION The combining therapy of CR and borneol induces a better improvement than monotherapy of CR in a region-specific manner,which is closely related to the dose of borneol.
