梅花鹿骨粉营养成分和抗骨质疏松活性研究 被引量:8

摘要 为探究梅花鹿骨粉的营养成分及对去卵巢大鼠骨质疏松症的治疗作用,对梅花鹿骨粉的营养成分:氨基酸、常量及微量元素和脂肪酸的含量进行了测定,并对梅花鹿骨粉抗骨质疏松活性进行了试验。将60只大鼠随机分为6组,每组10只,分别为假手术组、模型对照组、阳性对照组及梅花鹿骨粉高剂量组(600 mg/kg)、中剂量组(300 mg/kg)、低剂量组(150 mg/kg)。采用切除双侧卵巢法建立骨质疏松模型。各组从造模4周后,每天灌胃给药1次,连续90 d。各组大鼠处死前1天,分别用代谢笼收集24 h尿液标本,末次给药后1 h断头处死。检测骨密度、血钙、血磷、碱性磷酸酶及尿磷等生化指标。结果表明:梅花鹿骨粉中检出17种氨基酸,其中7种必需氨基酸占总氨基酸的20.69%;脂肪酸成分共检出9种,其中包括6种不饱和脂肪酸,占脂肪酸检出总量的48.78%;在常量和微量元素中,Ca和P的比值为2.3∶1,同时富含Fe和Zn。梅花鹿骨粉3个剂量组大鼠血中钙浓度明显升高,血中碱性磷酸酶活性明显降低。梅花鹿骨粉300 mg/kg剂量组和600 mg/kg剂量组中血磷浓度明显降低,而150 mg/kg剂量组无显著差异。梅花鹿骨粉300 mg/kg剂量组和600 mg/kg剂量组中尿钙/尿肌酐比值明显降低,骨密度明显升高,150 mg/kg剂量组无显著差异;3个剂量组大鼠尿中磷浓度都明显降低。结果显示,梅花鹿骨粉对去卵巢骨质疏松大鼠有良好的治疗作用。 In order to investigate the nutritional components of sika deer bone meal and its therapeutic effect on osteoporosis in ovariectomized rats, the content of the nutritional components in the sika deer bone meal was determined, including amino acids, macro and trace elements and fatty acids, and the anti osteoporosis activity of sika deer bone meal was tested. Sixty rats were randomly divided into 6 groups (n= 10), sham operation group, model control group, positive control group and high dosage group of sika deer bone meal (600 mg/kg) , middle dose group (300 mg/kg), low dose group ( 150 mg/kg). Resection of bilateral ovaries method was adopted to establish the model of osteoporosis. Four weeks after modeling, the rats were filled stomach 1 time a day for 90 days. One day before the rats were killed, the urine samples were collected in the metabolic cages for 24 hours, 1 hours after the last administration, the rats were slaughtered. Detection of rat bone density, serum calcium, serum phosphorus, alkaline phosphatase and urine phosphorus and other biochemical indi- cators were carried out. Results showed that bone meal of sika deer contained 17 amino acids and 7 essential amino acids accounted for 20.69% of total amino acids. Nine fatty acids were identified from bone meal of sika deer,including 6 unsaturated fatty acids with a relative content 48.78%. Ca and P ratio was 2.3 : 1, Fe and Zn as relatively abundant trace element were detected. Blood calcium concentration of rats in the group fed three dose of sika deer bone meal increased significantly, serum alkaline phosphatase activity decreased obviously. The blood phosphorus concentration of rats in the group fed sika deer bone powder 300 mg/kg dose and 600 mg/kg dose was significantly lower, and that 150 mg/kg dose had no significant difference. The urinary calcium/creatinine ratio of rats in the groups fed 300 mg/kg and 600 mg/kg dose of sika deer bone powder decreased significantly, bone mineral density increased significantly, had no significant difference in 150 mg/kg dose group. The urine phosphorus concentrations of rats in three dose groups were significantly lower. This finding indicated that bone meal of sika deer had significant effects on ovariectomized osteoporosis rats.
