摘要
目的:探讨p27KIP1与肝细胞癌的发生发展及其凋亡的关系.方法:利用免疫组织化学方法检测52例肝细胞肝癌组织中p27KIP1的表达,同时采用DNA缺口末端标记技术(TUNEL)检测凋亡细胞.结果:p27KIP1在癌旁肝组织中的阳性细胞指数(10.7±7.6)明显高于在癌组织中的阳性细胞指数(1.5±0.9,P<0.01).并且在包膜受侵犯者(1.4±0.76vs1.8±0.3,P<0.01)、肝内转移者(1.2±0.2vs1.5±0.4,P<0.05)及低分化的肝癌组织(1.9±0.7vs1.4±0.4,P<0.01)中p27KIP1蛋白表达明显降低.进一步的研究发现,肝癌组织中高AI(apoptosisindex)组的p27KIP1阳性细胞指数(1.7±0.6)明显高于低AI组(1.4±0.3,P<0.05).结结论论:p27KIP1基因的低表达与HCC的浸润、转移及恶性程度有着密切的联系.并且肝癌细胞的凋亡与p27KIP1基因的表达有关.
AIM: To study the correlation between expression of p27KIP1,progression and apoptosis in hepatocellular carcinoma(HCC).METHODS:Expression of p27KIP1 was detected by immu-nohistochemical method in 52 cases of HCC. And apoptosiswas examined by terminal deoxynucleotidyl transferase-mediated nick end labeling (TUNEL) technique.RESULTS:p27KIP1 LI was significantly higher in tumor-sur-rounding tissues than that in tumor tissues(10.7 ± 7.6 vs1.5±0.9, P <0.01). And HCC with intrahepatic metastasis,infiltration and poor-differentiation had significantly lowerp27KIP1 LI than those without intrahepatic metastasis(1.2±0.2 vs 1.5±0.4, P <0.05), no infiltration(1.4±0.76 vs 1.8±0.3, P <0.01) and well-differentiation (1.9± 0.7 vs 1.4±0.4, P <0.01). Moreover, the expression of p27KIP1 in highAI tumors was higher than that in low AI tumors (1.7± 0.6 vs 1.4 ± 0.3, P <0.05).CONCLUSION: p27KIP1 expression level in HCC is closelyassociated with the differentiation, infiltration and metastasis,and relates to its apoptosis.
