人卵巢颗粒细胞的体外分离培养方法改进 被引量:1

摘要 目的建立高效稳定的人卵巢颗粒细胞的体外分离培养方法。方法收集体外受精-胚胎移植取卵时的卵泡液,查阅近几年文献中人卵巢颗粒细胞的分离纯化方法,对沉淀法和梯度密度离心法进行改进,用改进了的沉淀法和密度梯度离心法相结合对卵巢颗粒细胞进行分离纯化,选择合适的消化时间,简化分离纯化步骤从而减少对颗粒细胞的损伤。结果分离出的人卵巢颗粒细胞生长状态良好,存活率高,收获的细胞数量多,后续生长稳定。结论建立了人卵巢颗粒细胞体外培养的稳定模型,为颗粒细胞的体外研究奠定良好的基础。 objective: To establish an effective method tbr isolating and culturing human ovarian granulosacell in vitro. Method: Follicular fluid was collected from women undergoing IVF procedures. Methods of isolation and purification of human ovarian granulosa cells in recent years were reviewed, Which were then made an improvement, as well an innovation of method combination to treat the Follicular fluid. Results: The isolated ovarian granulosa ceils showed a good growth, high survival rate, large number of harvested cells and stable subsequent growth. Conclusion: A stable model of human granulosa cells cultured in vitro has been established, which lays a good foundation for in vitro study ofgranulosa cells.
