瘦素在围着床期小鼠子宫组织中的表达 被引量:22

摘要 目的:探讨瘦素在围着床期小鼠子宫组织中的表达规律。方法:分别取妊娠d1-6NIH小鼠子宫,提取总RNA,检测子宫以及胚胎着床点和非着床点瘦素mRNA表达规律;同时石蜡包埋、切片,采用免疫组化试验,分别检测妊娠d1-6子宫瘦素蛋白的表达规律。结果:妊娠d1-6小鼠子宫组织均有瘦素mRNA和蛋白的表达。瘦素mRNA呈规律性表达,d1、d2表达微弱,d3开始增强,d4达到高峰,d5、d6仍维持在一定水平;另妊娠d5或d6,着床点瘦素mRNA水平比非着床点显著增强。瘦素蛋白主要定位于腔上皮和腺上皮。结论:小鼠胚胎围着床期,瘦素在子宫组织呈动态表达。提示瘦素可能与胚胎发育、胚胎着床的进程有关。 Objective: To investigate the effect of the Traditional Chinese Medicine Erzhi Tiangui Granules on embryo quality in patients undergoing controlled ovarian hyperstimulation. Method: Sixty-six infertile patients with tubal factors undergoing IVF-ET were divided into two groups at random, 33 cases in the study group treated with Erzhi Tiangui Granules +GnRHa/FSH/hCG, and 33 cases in control group were treated with GnRHa/ FSH/hCG. Concentration of IL-1β and IL-6 in follicular fluid on the day of retrieving oocytes were detected by ELISA, and embryo grade was evaluated on the day of embryo transfer. Results: Follicular fluid IL-1β concentration and embryo grade were significantly highter in experimental group than in control group (P〈0.05, P〈0.01). Follicular fluid IL-1β concentration was in positive correlation with the embryo grade (P〈0.01). Conclusion: Traditional Chinese Medicine for tonifying kidney can promote embryo maturation by regulating IL-1β concentration in follicular fluid.
