摘要
在国内外首次证明了动脉壁心钠素(ANF)的存在,并探讨了体外培养的大鼠主动脉平滑肌细胞(ASMC)合成和分泌ANF的能力及可能的分泌机制。结果表明,体外培养的ASMC能合成和分泌ANF。卒中易感型自发性高血压大鼠(SHRsp)的ASMC及其培养液内ANF含量低于,而ASMC内肾素活性(RA)高于WKY大鼠。本结果对探讨高血压发病的血管机制有重要意义。
Atrial natriuretic factor (ANF) was found to e present in the aortic wall for the f(?)st time. HPLC and RIA of ANF of cultured ASMC and medium revealed that ANF in the cells was ANF(31-126) in amounts of 0.82±0.03(SHBsp) and 1.04±0.03ng/10~3 cells (WKY), while that in the medium was ANF (99-126) with contents of 0.40±0.06 and 0.60±0.06ng/10~(?) cells respectively. The contents of ANF in both ASMC and medium of SHBsp were significantly lower than those of WKY rats. On the contrary, both renin activity and angiotensin Ⅱ concentration in ASMC of SHBsp were significantly higher as compared with those of WKY rats. In addition, immunocytochemistry showed positive ANF staining in cultured ASMC of both strains. The results suggest that ANF can be synthesized and secreted by cultured ASMC of rats.
