摘要
以麦角甾醇为测定目标,建立测定山茶油中麦角甾醇的高效液相色谱法。样品经皂化提取后,采用高效液相色谱法进行测定。色谱条件为:C18柱(4.6 mm×250 mm,5μm);流动相为纯甲醇;流速1.0 m L/min;紫外检测器:波长282 nm;进样量20μL;外标法定量。结果表明:麦角甾醇线性回归方程为Y=31.173X+0.3008(X为标物浓度,mg/L),R2为0.99999,在0.50~10.0mg/L范围内线性良好,方法回收率为94.5%~102%,RSD为3.56%,检出限为1.0 mg/kg。本方法测定茶油中麦角甾醇含量准确、重复性好、灵敏度高。
A new method for determination of ergosterol in camellia oil by reversed - phase high performance liquid chromatography. Peanuts were analyzed by high performance liquid chromatography after saponification with KOH - ethanol solution. The separation of ergosterol was conducted on a C18 column (4.6 mm × 250 mm, 5μm ) with the mobile phase of methanol. The flow rate was 1.0 ml-/min. The detection wavelength was 282 nmand injection volume was 20μL. The egosterol was quantified by the external standard method. The linear regression equation was Y = 31. 173X + 0.3008 with the range of 0.50 -10.0mg/L. The correlation coefficients was 0.9999. The average recoveries of samples was in the range of 94.5% -102% and RSD was 3.56%. The determination of limit wasl. 0mg/kg. The results indicate that the method is accurate , reliable and reproducibility in the analysis of camellia oil.
