摘要
目的:初步探讨健脾化瘀解毒中药复方胃痞灵(WPL)抗慢性萎缩性胃炎(CAG)的效应。方法:大鼠随机分为空白组、模型组、胃复春组(WFC,0.39g/kg)和WPL组(9g/kg)各10只,复制氨水所致CAG大鼠模型,连续给药8周后测血清SOD活性和MDA、NO含量,取胃液测胃蛋白酶和0.5mL胃液中游离胃酸含量,取胃小弯体窦交界胃窦,观察胃黏膜病理形态学变化并测定胃黏膜层和基底膜平均厚度。结果:模型组大鼠较WFC和WPL组胃黏膜上皮固有腺萎缩、部分腺体假幽门腺化生、黏膜微血管扩张充血、基底膜明显增厚:游离胃酸含量、胃蛋白酶活性、SOD、NO和胃黏膜平均厚度较空白组均有显著性降低(P<0.01),MDA有显著性增加(P<0.01):WPL组大鼠游离胃酸含量、胃蛋白酶活性、SOD、NO和胃黏膜平均厚度较模型组均有显著性增加(P<0.01),MDA有显著性减少(P<0.05)。结论:WPL可修复受损胃黏膜,逆转胃黏膜萎缩、肠上皮化生,恢复胃酸和胃蛋白酶分泌功能,其机制可能与提高SOD活性和NO含量同时降低MDA含量有关。
Objective: To investigate the effect of the Chinese herbal compound for tonifying spleen and removing blood-stasis and detoxifica- tion-Weipiling (WPL) against chronic atrophic gastritis (CAG). Methods: Rats were randomly divided into control group, model group, Weifuchun group (WFC, 0.39g/kg) and Weipiling group (WPL, 9g/kg) which had 10 cases in each group, CAG model rats induced by ammonia were duplicated and administered for 8 successive weeks then detected their serum SOD activity and contents of MDA and NO, gastric juice was taken to measure the contents of pepsin and free gastric acid in 0.5mL gastric juice, and observed the change of gastric mucosa pathology in gastric antrum that at the edge of corpora ventriculi and sinuses ventriculi and measured the average thickness of gastric mucosa and basement membrane. R^sults: There were more intrinsic glandular atrophy of gastric epithelium, pseudopyloric metaplasia in partial glandular, mucosal microvascular dilatation and conges- tion and basement membrane markedly thickened in model group than WFC and WPL group; compared with control group, the pH of free gastric juice, pepsin activity, SOD, NO and thickness of gastric mucosa of model group were significantly decreased (P〈0.01) while MDA significantly in- creased (P〈0.01); compared with model group, the pH of free gastric juice, pepsin activity, SOD, NO and thickness of gastric mucosa of WPL group were significantly increased (P〈0.01) while MDA significantly decreased (P〈0.05). Conclusion: WPL can effectively repair the injured gastric muco- sa so as to reverse the gastric mucosal atrophy and intestinal metaplasia, restore the secretion function of gastric acid and pepsin in rats, which mecha- nism may be associated with SOD activity and NO content increasing and MDA content decreasing.
