Pin1在盐酸异丙肾上腺素诱导的心肌纤维化大鼠中的表达

摘要 目的探讨肽脯氨酰顺反异构酶(Pin1)在盐酸异丙肾上腺素(ISO)诱导的心肌纤维化大鼠中的表达。方法 27只SD大鼠随机分为对照组(10只)、实验组(17只),其中实验组腹腔注射ISO,对照组注射等体积生理盐水,持续用药21 d,最终对照组和实验组各有10只大鼠纳入研究。超声心动图测定两组大鼠射血分数、短轴缩短率、左室收缩末期内径和左室舒张末期内径等相关心功能指标后处死取材,分别行苏木精-伊红染色(HE染色)、苦味酸-天狼猩红染色(PSR染色)观察心肌纤维化情况;实时荧光定量PCR技术检测Pin1、Ⅰ型胶原蛋白、Ⅲ型胶原蛋白mRNA表达水平。结果病理形态学观察显示实验组大鼠心肌纤维化程度较高。实验组射血分数、短轴缩短率均低于对照组,Ⅰ型胶原蛋白、Ⅲ型胶原蛋白、Pin1 mRNA表达水平均高于对照组(P<0.05)。结论 Pin1或参与了ISO诱导的心肌纤维化,可能是心肌纤维化的潜在治疗靶点。 Objective To investigate the expression of peptidyl-prolyl cis-trans isomerase,Pinl ,in cardiac fibrosis rats induced by isoproterenol(ISO). Methods Twenty-seven Sprague-Dawley rats were randomly divided into control group (n = 10 ) and experimental group( n = 17 ). The experimental group was given ISO through intraperitoneal injection ,while the control group was given equal volumes of normal saline by the same administrated method, and the continuous medication was used for 21 days in the two groups. Finally, 10 rats were enrolled in each group. Echocardiogram was used to determine the relevant cardiac indices including ejection fraction, shortening fraction,left ventricular end-systolic diameter and left ventricular end-diastolic diameter, then the rats were sacrificed for specimens. The myocardial fibrosis was observed by hematoxylin-eosin staining(HE staining) and picric acid-Sirius red staining( PSR staining) separately. The mRNA expression levels of Pinl ,collagen Ⅰ and collagen Ⅲ were detected by real-time fluorescent quantitative PCR. Results Pathological morphological observation showed that there was a higher degree of myocardial fibrosis in the experimental group. Compared with the control group, the ejection fraction and shortening fraction were lower while the mRNA expression levels of Pinl, collagen Ⅰ and collagen Ⅲ were higher in the experimental group(P 〈 0.05). Conclusion Pin1 might play a role in cardiac fibrosis induced by ISO, and might be a potential therapeutic target for cardiac fibrosis.
