摘要
目的:制备模拟人类发病过程的大鼠2型糖尿病心肌病模型,探讨加味生脉补心丹组对糖尿病心肌病大鼠的血糖、血脂及脂联素水平的影响及分子机制。方法:用雄性5周龄SD大鼠40只,随机分组如下:正常对照组(n=8),高糖高脂组(n=8),模型组(n=24),高糖高脂喂养4w造成胰岛素抵抗后,正常对照组和高糖高脂对照组分组予以普食和高糖高脂饲料喂养。模型组于第5周予以腹腔注射2次1%小剂量STZ,2次之间间隔48h,糖尿病造模成功后,随机继续高糖高脂喂养22w。测定胰岛素、脂联素、糖化血红蛋白、空腹血糖、TG、TC、HDLC、LDLC。结果:中药组和西药组的外在表现均优于模型组和正常组;TG、TC、HDLC的值低于模型组,HDLC高于模型组;胰岛素水平高于模型组,说明存在模型组高胰岛素血症,存在胰岛素抵抗;脂联素水平高于模型组。结论:加味生脉补心丹治疗糖尿病心肌病机制可能改善糖脂代谢与升高脂联素水平有关。
Objective: To prepare simulate human pathogenesis of type 2 diabetic cardiomyopathy rat model of Jiawei Shengmai Buxin Dan group of diabetic cardiomyopathy rats blood glucose, blood lipids and adiponectin and molecular mechanisms of influence.Methods: The 5-week-old male SD rats were randomly divided as follows: normal control group (n=8), high glucose and high fat group (n=8), model group (n=24), high-sugar high-fat diet after 4w cause insulin resistance, normal control group and high glucose and lipid control packet to be normal diet and high-sugar high-fat diet.Model group to be in the first five weeks by intraperitoneal injection of 2 times 1% low-dose STZ, the interval between 48h 2 times, after successful modeling diabetes randomized to continue high-sugar high-fat diet 22w.Determination of insulin, adiponectin, glycated hemoglobin, fasting plasma glucose, TG, TC, HDLC, LDLC.Results:1 Medicines and external manifestations of Western medicine group were better than the model group and the normal group;2 value TG, TC, HDLC is lower than the model group, HDLC higher than the model group;3 insulin levels higher than the model group,indicating the presence of model group hyperinsulinemia, insulin resistance;4 adiponectin levels than the model group;Conclusion: Jiawei Shengmai Bu Xin Dan treating diabetic cardiomyopathy mechanism may improve glucose and lipid metabolism and elevated adiponectin levels.
