摘要
目的探讨脊髓自噬功能与糖尿病神经病理性疼痛(DNF)的相关性。方法选取STZ制备的糖尿病模型,随机分为制模后3 d组、制模后1周组、制模后2周组、制模后3周组及制模后4周组,每组20只。同期选取5组健康大鼠分为对照1~5组(Con 1~5),每组20只,注射柠檬酸缓冲液。比较各组血糖、机械疼痛阈值、热刺疼痛阈值,以及脊髓自噬功能和脊髓补体C3的表达。结果制模后各组血糖均高于同期对应的Con组(P<0.05),除制模后3 d组外,其余各组机械疼痛阈值低于同期对应的Con组(P<0.05)。制模后3周组、制模后4周组热刺激疼痛阈值也低于同期对应的Con组(P<0.05)。制模后2周组、制模后3周组和制模后4周组C3 mRNA、补体C3、Beclin-Ⅰ和LC3-Ⅱ表达均高于同期对应的Con组(P<0.05)。制模后3 d组、制模后1周组、制模后2周组、制模后3周组和制模后4周组P62蛋白表达均低于同期对应的Con组(P<0.05)。结论大鼠脊髓补体C3、Beclin-Ⅰ、LC3-Ⅱ、P62激活提示脊髓自噬功能激活;脊髓自噬功能激活可能参与了DNP发生发展。
Objective To investigate the correlation between spinal cord autophagy and diabetic neuropathic pain(DNP).Methods STZ-induced diabetic rats were randomly divided into 5 groups:DM-3d group,DM-1w group,DM-2w group,DM-3w group and DM-4w group,with 20 rats in each group.Heath rats were correspondingly selected as 5 control groups(Con1-5) also with 20 rats in each group and treated with citrate buffer solution.The blood glucose,mechanical pain threshold,thermal stimulation pain threshold,the autophagy and the expression of complement C3 in the spinal cord were compared.Results The levels of blood glucose in each DM group were higher than those in corresponding Con groups(P〈0.05).The mechanical and thermal stimulation pain thresholds decreased in DM groups(P〈0.05).The expressions of C3 mRN A,complement C3,BeclinT and LC3- Ⅱ were higher in DM-2w,DM-3w and DM-4w group than those in corresponding Con groups(P〈0.05).The expressions of P62 protein decreased in DM-3d,DM-1w,DM-2w,DM-3w and DM-4w group(P〈0.05).Conclusion The expressions of C3,Beclin-I,LC3-Ⅱ and P62 may be related to the activation of spinal cord autophagy that may be involved in the development of DNP.
