摘要
目的:研究清眩降压汤对自发性高血压模型大鼠(SHR)的血压控制和大血管重构的干预作用。方法:20只SHR按照随机数字大小排序分为清眩降压汤组(治疗组)和模型对照组(对照组),另设Wistar大鼠10只作为正常血压对照组。干预期间,每周测量大鼠的血压、体质量,实验8周结束后取血管用于病理组织学观察,测量血管中膜厚度(MT)和腔径(LD),计算LD/MT的比值,检测Bax、Bcl-2的基因蛋白表达水平。结果:治疗组大鼠血压低于对照组(P<0.05);清眩降压汤能够减小血管中膜厚度和LD/MT的比值,与对照组比较差异有统计学意义(P<0.05);但2组对动脉腔径影响差异无统计学意义(P>0.05)。治疗组血管平滑肌细胞Bcl-2的表达和Bcl-2/Bax的比值较对照组低,差异有统计学意义(P<0.05)。结论:清眩降压汤可以控制SHR血压的进一步升高,抑制血管平滑肌细胞增殖,改善血压升高导致的动脉血管重构。
Objective:To discuss the therapeutical effect of Qingxuan Jiangya decoction(QXJYD) on spontaneously hypertensive rats(SHR). Methods:By using SHR as hypertension model,rats were randomized into two groups:SHR+QXJYD group and SHR group,and ten Wistar-Kyoto rats were set as WKY group. During experimental period systolic blood pressure(SBP) and body weight were measured weekly. After eight weeks with the treatment of QXJYD,the aortas tissue was collected for histopathologic examination. The media thickness(MT) and lumen diameter(LD) were measured,ratio of lumen diameter / media thickness(LD/MT)in the carotid arteries were calculated,the gene and protein expression levels of Bax and Bcl-2 were detected. Results:SBP in SHR+QXJYD group was obviously lower than that in SHR group(P〈0.05). QXJYD significantly decreased MT and ratio of LD/MT in the carotid arteries(P〈0.05). QXJYD could markedly reverse vascular remodeling,and had no significant influence on lumen diameter of artery(P〉0.05). A significant reduced expression of anti-apoptotic Bcl-2 proteins and ratio of Bcl-2/Bax were found in SHR+QXJYD group compared with those in SHR group(P〈0.05). Conclusion :QXJYD can reverse vascular remodeling by controlling the blood pressure of SHR and inducing proliferation of vascular smooth muscle cells(VSMCs) in vivo.
