Endothelin (ET) is a recently discovered vasoaotive peptide. It has been shown to have potent vasoconstriotive effects in vivo and in vitro. The present study in anaesthetized rats was to investigate the effects of ET on blood pressure and renal function including glomerular filtration rate (GFR), urine flow (UV), renal sodium excretion (UNa+V), renal potassium excretion (UK+V) and fractional sodium excretion (FENa+) by intravenous bolus injection of ET. Results showed that ET caused a biphasic arterial blood pressure response with a marked lowering at the every start, and a rapid elevation later. It also caused significant decrement in GFR (P<0.001), UV(P<0.001), UNa+·V(P<0.05) and UK+·V(P<0.01). FENa+ was not significantly changed during the test. The findings cited above indicate that ET has a marked pressor effect, accompanied by a significant decrease in GFR and renal electrolyte excretion.
