摘要
青霉菌 (Penicilium sp.)发酵液经 2 0 %~ 70 %硫酸铵分级沉淀 ,DEAE- Cellulose离子交换层析 ,Sephadex G-10 0凝胶过滤层析 ,得到聚丙烯酰胺凝胶电泳均一的壳聚糖酶 ,比活力 10 2 0 u/ m g,纯化 182倍。其分子量为 30 k D,酶的最适温度和 p H分别为 5 0°C和 5 .0 ,金属离子 Mg2 +和 Ca2 +对酶活力有一定的促进作用 ,重金属离子 Cu2 +、Ni2 + 和 Zn2 + 对酶有较强的抑制作用。该壳聚糖酶具有很强的底物专一性 ,动力学参数 Km 值为 2 .6 0 1g/ L。
Chitosanase was isolated and purified from the supernatant of fermentation broth of Penicillum sp.. The purification procedure involved ammonium sulfate fraction, ion exchange chromato graphy on DEAE Cellulose column, gel filtration chromatography on Sephadex G 100 column. The purified enzyme was homogeneous and estimated 30 kD on SDS PAGE. The optimal temperature for the enzyme was 50 °C and optimal pH was 5.0. Metal ions of Mg 2+ and Ca 2+ can activate the enzymatic reaction, but heavy metal ions of Zn 2+ ,Ni 2+ and Cu 2+ inhibited the enzyme activity. The chitosanase showed substrate specificity. Kinetic parameter K m was 2.601 g/L.
