白芍总苷脂质体诱导胃癌BGC-823细胞凋亡的实验研究 被引量:13

摘要 目的探讨白芍总苷脂质体对人胃癌BGC-823细胞凋亡的诱导作用。方法 CCK-8试剂盒检测白芍总苷脂质体对BGC-823细胞增殖的影响,HE染色、Hochest 33342/PI双染和透射电子显微镜观察BGC-823细胞凋亡的形态学变化,Western blotting检测细胞凋亡相关因子Bcl-2和Bax蛋白的表达情况。结果白芍总苷脂质体能明显抑制BGC-823细胞的增殖,呈现一定的剂量依赖关系(P<0.05)。白芍总苷脂质体干预BGC-823细胞48 h后,凋亡形态学观察均可见细胞核固缩、碎裂和凋亡小体等凋亡特征改变,Western blotting检测Bcl-2蛋白表达减少,Bax表达增多,Bcl-2/Bax比值降低。结论白芍总苷脂质体明显诱导胃癌BGC-823细胞凋亡,抑制其增殖,其机制可能与下调Bcl-2蛋白、上调Bax蛋白及降低Bcl-2/Bax比值有关。 OBJECTIVE To study the apoptosis of human gastric cancer cell line BGC-823 induced by liposome of total gluco- sides from paeonia (TGP liposome ). METHODS The inhibition of cell proliferation was determined by Cell Counting Kit-8 as- say. The morphological changes of the apoptosis cells were observed by HE staining, Hoechst 33258 fluorescent staining, transmission e- lectron microscope. Changes of apoptosis related factor Bcl-2 and Bax protein expression were detected by Western blot. RESULTS TGP liposome significantly inhibited proliferation of BGC-823 cells in a dose-dependent manner( P 〈 0. 05 ). GC-823 cells induced by TGP liposome for 48 h was observed on morphological changes such as karyopyknosis and karyorrhexis and apoptotic bodies. Bel-2 ex- pression was reduced, but Bax expression was increased, so the ratio of Bcl-2 /Bax decreased. CONCLUSION TGP liposome can induce the apoptosis of BGC-823 cells and inhibit its proliferation, its mechanism may be related to down-regulating protein expression of Bel-2, up-regulating protein expression of Bax, reducing Bcl-2/Bax value.
