响应面法优化山茱萸籽蛋白制备工艺条件的研究 被引量:3

Optimization of Protein Extraction from Cornus officinalis Seed Meal by Response Surface Methodology

摘要 采用酶碱两步法制备山茱萸籽蛋白,在碱提酸沉法的基础上,用糖化酶辅助提取制备山茱萸籽蛋白。对制备工艺中液料比、酶用量、酶作用时间和酶作用pH分别进行了单因素试验,在单因素试验的基础上,采用响应曲面法(RSM)优化制备工艺条件,结果表明,影响山茱萸籽蛋白的蛋白得率的4个主要因素的主次关系为:酶用量>酶作用时间>料液比>酶作用pH。最佳工艺条件为:液料比13︰1 mL/g,酶用量0.04%,酶作用时间44 min,酶作用pH 4.4。验证试验表明,在此条件下的山茱萸籽蛋白得率为90.25%,接近分离蛋白的标准。 Use alkali dissolution and enzyme hydrolysis to get Comus officinalis seeds protein. On the base of alkali extraction and acid precipitation, glucoamylase was used to assisted during the Comus officinalis seeds protein extraction. The experiments were carded out to test the yield of Comus o^cinalis seeds protein affected by different factors which including the ratio of liquid to material, enzyme dosage, enzyme action time and pH. Based on single factor test results, response surface methods were used to optimize the method of alkali dissolution and enzyme hydrolysis to get Comus officinalis seeds protein. The results showed the impaction strength of the four main factors of Comus officinalis seeds protein extraction were as follows: enzyme do sage〉.enzyme action time 〉ratio of liquid to material〉pH. Optimal conditions for the preparation by alkali dissolution and enzyme hydrolysis were that liquid-solid ratio was 13 :1 mL/g, the amount of enzyme was 0.04%, enzyme action time was 44 min, pH of enzymatic reaction was 4.4. Verification tests were made and yield of protein under these conditions was 90.25%, which was close to the standard of protein isolate.
