粗毛纤孔菌固体发酵菌粉对H_(22)荷瘤小鼠抗肿瘤作用研究 被引量:20

Antitumor Effect of Solid State Fermentation Powder of Inonotus hispidus on H_(22) Bearing Mice

摘要 目的:探讨粗毛纤孔菌固体发酵菌粉的抗肿瘤作用及其机制。方法:将90只小鼠分为9组,分别为正常对照组、模型组、环磷酰胺阳性对照组及粗毛纤孔菌玉米发酵菌粉高、中、低剂量组和高粱发酵菌粉高、中、低剂量组。H_(22)荷瘤小鼠接种后,连续给予相应药物11 d,然后对H_(22)荷瘤小鼠的抑瘤率、体质量、胸腺指数、脾脏指数及肝脏指数进行测定,对各组小鼠的肿瘤组织进行组织病理学检查,并对肝组织及血清中的MDA、SOD、CAT及GSHPx水平进行测定,同时测定血清中细胞因子IL-2、VEGF及IFN-γ含量。结果:粗毛纤孔菌发酵菌粉可增加小鼠体质量,提高胸腺指数、脾脏指数及肝脏指数并抑制肿瘤生长;经组织病理学观察,肿瘤坏死细胞明显增加;并可升高血清及肝组织SOD、CAT及GSH-Px水平,降低MDA水平;升高血清IL-2和IFN-γ水平,降低VEGF水平。结论:粗毛纤孔菌固体发酵菌粉具有抗肿瘤作用,其机制可能与机体抗氧化、提高机体免疫力及抑制肿瘤组织血管生成有关。 Objective :To study the inhibitory effect and the mechanism of solid fermentation powder(SFP) of Inonotus hispidus on H22 bearing mice. Methods:The H22 transplanted models of mice were established by H22 ascites tumor. 90 mice were divided into nine groups as control, model, cytoxan ( CTX,20 mg/kg) positive control, high-, mid-, low-dose ( 1 000,750, and 500 mg/kg) of corn SFP of Inonotus hispidus, and high-, mid-, low-dose ( 1 000,750, and 500 mg/kg ) of sorghum SFP of Inonotus hispidus. On the day after being inoculated with H22 tumor cells, mice were given SFP of lnonotus hispidus once daily for consecutive 11 d. And then the increaseing of the tunmr inhibitory rate, body weight, thymus, spleen, and liver indexes were calculated ; and the histopathologic changes of tumor of He2 bearing mice were observed. The activities of MDA,SOD and CAT,the contents of GSH-Px in liver homogenates and blood serum,and the number of IL-2,VEGF,and IFN-y in blood serum were detected. Results:The treatment with SFP of Inonotus hispidus significantly increased the body weight,thymus index, spleen,and liver indexes. The histopathologic study showed that the necrosis in H22 cells was induced, and the hepatic SOD, CAT, GSH-Px activities were increased,the MDA in liver homogenates and blood serum were decreased, the serum levels of IL-2 and IFN-γ were elevated, and tbe VEGF level was reduced. Conclusion:SFP of Inonotus hispidus have an antitu- mor effect on H22 bearing mice,and the possible mechanism may be due to its antioxidant activity,improving immunofunction, and anti- angiogenetie action.
