摘要
目的:考察不同产地粉葛中葛根素、3'-甲氧基葛根素、大豆苷、大豆苷元的含量,为制定粉葛药材质量标准提供参考依据。方法:采用RP-HPLC法,色谱柱Hypersil C_(18)色谱柱(4.6 mm×250 mm,5μm);流动相:乙腈-0.2%磷酸,梯度洗脱;流速1.0 ml/min,柱温30℃,检测波长250 nm。结果:葛根素、3'-甲氧基葛根素、大豆苷、大豆苷元的线性范围分别为12.4~620ng、1.52~76ng、2.4~120ng、3.28~164 ng,四种成分的平均加样回收率分别为101.1%、99.86%、101.78%、103.39%,RSD分别为1.43%、1.29%、0.95%、1.21%。结论:该方法简便快速,重复性良好,结果准确可靠,可用于不同产地粉葛药材中四种异黄酮类成分的含量测定。
Objective To determine the content of puerarin, 3' - methoxy puerarin, daidzin, daidzein in different habitats of radix puerariae thomsonii, and provide reference for establishing its quality standards. Methods HPLC determination was performed on a Hypersil C18 column (4. 6 mm ×250 mm, 5 μm) and detected at 250 nm. The mobile phase was consisted of Acetonitrile and 0. 2% Phosphate acid solution with gradient elution. The flow rate was 1.0 ml · min^-1 and column temperature was kept at 30℃. Results The method was proved to be linear in the ranges of 12. 4 -620, 1.52 -76, 2. 4 - 120, 3.28 - 164 ng for the four isoflavonoids, puerarin, 3'-methoxy puerarin, daidzin, daidzein, respectively. The average recoveries were 101.1% (1.43%), 101.78% (0.95%), 103. 39% (1.21%), respectively. Conclusion Results obtained showed that the method is convenient, accurate and reliable, can be applied to the determination of the content of Radix Pueraria thomsonii from different sources in four isoflavonoids.
