摘要
目的 :探讨用高效液相色谱法定量测定葛根素的含量。方法 :色谱柱 大连SpherisorbC18( 10 μm ,4 6mm× 2 50mm) ;流动相 水 (用冰醋酸调至pH3 0 ) 甲醇 ( 80∶2 0 ) ;二极管阵列检测器 ,检测波长 2 50nm。结果 :葛根素与 3' 甲氧基葛根素完全分离。浓度线性范围为 2 0~ 180 μg/ml(r =0 9999) ,平均加样回收率为10 1 7% ,RSD =0 4 3% (n =5)。结论 :本法简便、准确 ,重现性好 ,可用于葛根素的质量控制。
Objective: To investigate a HPLC method for quantitative determining puerarin Method: The chromatographic conditions were as follow: Dalian Spherisorb C 18 (10μm,4 6mm×250mm),water(adjusted to pH=3 0 with glacial acetic acid) methanol (volume ratio 80∶20) as the mobile and a Waters 996 photodiode array detector with wavelengh at 250nm Result:Good separation was achieved between puerarin and 3' methoxy puerarin The calibration curve was linear within 20~180μg·ml -1 and the correlation coefficient was 0 9999 The average recovery rate and RSD were 101 7% and 0 43%(n=5) Conclusion:This method is simple,accurate and reproducible and can be used for the quality control of puerarin
