摘要
丝状真菌三孢布拉霉是一种最有潜力大规模生产番茄红素的微生物。该研究利用实验室选育获得的1株三孢布拉霉,对其发酵条件进行了优化。首先采用单因素试验对其培养基的碳源、氮源、油类添加物、接种量和正负菌比例进行了优化,确定玉米粉、黄豆粉、棉籽油、接种量10%和正负菌比例1∶5分别为最佳选择。随后采用正交设计对玉米粉、黄豆粉、棉籽油和阻断剂的浓度进行了试验,进行了极差分析和方差分析,确定了最佳组合。在此条件下进行验证试验获得番茄红素的产量为29.9 mg/g菌体干重。
The filamentous fungi Blakeslea trispora is one of the most promising microorganism for the large-scale production of lycopene. This study optimized the fermentation conditions of a strain of Blakeslea trispora obtained from laboratory by breeding. Firstly,the single factor experiments on carbon source,nitrogen source,oil additives,inoculation amount and the ratio of positive and negative mating type were carried out,and results showed that corn flour,soybean meal,cottonseed oil,10% for inoculation amount and 1∶ 5 for the ratio were the best choice. The concentration of corn flour,soybean meal,cottonseed oil and the blocking agent was tested by orthogonal design and the optimum combination was determined by range analysis and variance analysis. Under this condition,the yield of lycopene was 29. 9 mg / g.
