摘要
本试验旨在利用PCR-DGGE(变性梯度凝胶电泳)技术研究日粮中不同稻壳替代水平对19~28日龄扬州鹅肠道微生物区系的影响。选用19日龄扬州公鹅260只,随机分成4组,每组5个重复,每个重复13只。A组饲喂基础日粮,B、C、D组分别饲喂7%、14%、21%稻壳替代的基础日粮。在28日龄时采集各肠段内容物进行总DNA的提取,对细菌16S r RNA V3区用细菌通用引物进行PCR扩增,利用DGGE电泳技术分析肠道微生物区系。结果显示:不同稻壳替代水平对十二指肠、空肠、盲肠菌群条带总数量影响不明显,但共有条带存在一定差异;十二指肠、空肠各组间菌群相似性程度均较低,盲肠组间相似性程度最高。由此可见,不同程度的稻壳替代能够影响肠道菌群分布,其中对十二指肠、空肠影响较大,对盲肠影响较小。
This experiment was conducted to study the effects of different proportions of rice husk replaced diets on intestinal microflora of geese from 19 - 28 days by denaturing gradient gel electrophoresis.Two hundred and sixty 19-day- old healthy Yangzhou geese were randomly assigned into 4 groups with 5 replicates of 13 geese each.The control group (A) were fed with basic diet (A);groups B,C,D were fed with 7%, 14%,21% rice husk replaced diets,respectively.The genomic DNA was extracted from the content of various intestinal segments at the age of 28 days.The V3 variable region of bacterial 16S rRNA was amplified with universal primer.Denaturing gradient gel electrophoresis profiles of the amplicons were analyzed.The results showed that different proportions of rice husk replaced diets had no significant effects on the band quantity of microflora in duodenum jejunun,and cecum of geese ,the common bands were different.And different pro- portions of rice husk replaced diets had no significant effects on the microflora similarity degree in duodenum and je- junum, the similarity index in cecum was maximal.Therefore, different proportions of rice husk replaced diets mostly affect- ed intestinal microfiora of duodenum and jejunum and rarely affect the cecum.
