摘要
通过单因素摇瓶培养结合50 L发酵罐参数优化,对毕赤酵母基因工程菌生产β-葡萄糖苷酶发酵条件进行研究,确定毕赤酵母β-葡萄糖苷酶工程菌摇瓶最佳产酶条件:增值阶段最适初始p H5.5、接种量10%、装液量100 m L/500 m L,50 L发酵罐中试条件:诱导p H6.0、诱导时间96 h、甲醇浓度1.0%(v/v)、溶氧控制20%~30%,在此条件下,酶活力可达98.85 IU/m L。进一步优化,可为提高木质纤维素生产纤维乙醇过程中酶解得率提供技术支撑。
To improve the cellulose saccharity rate,the fermentation conditions of Pichia pastoris producing β–glucosidase were studied.By single factor test and parameter optimization for pilotscale,the pilot-scale process were acquired.The optimal shake flask conditions were that initial p H5.5,inoculums size 10%,the volume of liquid 100 m L/500 m L.The fermentation conditions were initial p H6.0,induction time 96 h,methanol concentration 1.0%(v/v),DO volume 20%~30%,β-glucosidase activity 98.85 IU/m L.For further optimization,the technique can be provided for improving the hydrolysis yield of lignocellulosic.
