摘要 目的动态观察Rab-1和ERK在裸鼠移植血管瘤中的表达,探讨其在血管瘤自然退化中的作用。方法动态观察移植瘤大小、形态变化,免疫组织化学检测Ra61、p-ERK1/2蛋白的分布及其表达情况,RT-PCR检测Raf-1mRNA、p-ERK1/2mRNA的表达情况。结果RaP1在增殖期移植血管瘤组织中的阳性表达率为100%,IOD值为89.39±9.67,Raf-1mRNA的相对定量为0.33±0.03;退化期移植血管瘤阳性表达率为55%,IOD值为60.73±9.66,Raf-1mRNA的相对定量为0.21±0.02,两者各项值比较,差异均有统计学意义(P〈0.01或P〈0.05)。p-ERK1/2在增殖期移植血管瘤组织中的阳性表达率为100%,IOD值为85.65±10.88,p-ERK1/2mRNA的相对定量为0.29±0.02;退化期移植血管瘤阳性表达率为63%,IOD值为49.78±11.20,p-ERK1/2mRNA的相对定量为0.20±0.02,两者各项值比较,差异均有统计学意义(P〈0.01或P〈0.05)。Raf-1蛋白与p-ERK1/2蛋白的表达量呈正相关关系(r=0.8550,P〈0.01),Raf-1mRNA与p-ERK1/2mRNA的相对定量呈正相关关系(r=0.8750,P〈0.01)。结论裸鼠移植的血管瘤中有Raf-1、p-ERK的表达;Ra51/ERK信号通路可能参与了血管瘤的发生发展过程。 Objective To establish a reliable model of human hemangioma in nude mice for observing the expressions of Raf-1 and ERK in grafts so as to explore the roles of Raf-1/ERK signaling pathway in the occurrence, development and remission of pediatric hemangioma. Methods The expressions of Ra&l and ERK in xenografted human hemangioma in proliferating and involuting phases were detected by hematoxylin-eosin staining, immunohistochemistry and RT-PCR (reverse transcription-polyrnerase chain reaction). Results The expression rates of Raf-1 were 100% and 55% in proliferating and involuting phases respectively (P〈0. 01 ) ; IOD 89. 39 ± 9. 67 and 60. 73 ±9. 66 (P〈0. 05) ; Raf-lmRNA 0. 33 ±0. 03 and 0. 21 ± 0. 02 respectively (P〈0. 05). The expression rates of ERK were 100% and 63% in proliferating and involuting phases (P〈0. 01 ); IOD 85.65 ± 10. 88 and 49. 78 ± 11.20 (P〈0. 05) ; p-ERK1/2mRNA 0. 29± 0. 02 and 0. 20 ±0. 02 (P〈0. 05). The protein expression levels of Raf-1 and p-ERK1/2 had a positive correlation (r = 0. 855 0, P〈0. 01). And the mRNA relative quantities of Raf-1 and ERK had a positive correlation (r = 0. 8750, P〈 0. 01). Conclusions Raf-1 and p-ERK protein and gene are expressed in xenografts of nude mice. And Raf-1/ERK signaling pathway may be involved in the occurrence of hemangioma.
