摘要
目的:探讨脑胶质瘤患者脑脊液中波形蛋白水平及其临床意义。方法采用酶联免疫吸附试验( ELISA)法检测50例脑胶质瘤患者(胶质瘤组,其中WHOⅠ~Ⅱ级22例、WHOⅢ~Ⅳ级28例)及脑外伤、脑出血患者23例(对照组)的脑脊液中波形蛋白水平;Western Blot法检测WHOⅠ~Ⅱ级组、WHOⅢ~Ⅳ级组及对照组脑脊液游离细胞中波形蛋白的表达强度;Transwell法分析胶质瘤组患者脑脊液肿瘤细胞的迁移能力。结果脑脊液波形蛋白水平从高到低依次为WHOⅢ~Ⅳ级组、WHOⅠ~Ⅱ级组、对照组( P<0.05);Spearman相关分析显示脑脊液波形蛋白水平与胶质瘤恶性程度呈正相关( r=0.783, P<0.001);WHOⅠ~Ⅱ级组与WHOⅢ~Ⅳ级组脑脊液游离细胞中波形蛋白表达均较对照组显著增高,且WHOⅢ~Ⅳ级组升高较WHOⅠ~Ⅱ级组更为显著;WHOⅢ~Ⅳ级组脑脊液肿瘤细胞发生迁移的细胞数为(91.83±3.55)个,明显多于WHOⅠ~Ⅱ级组(51.37±3.35)个(P<0.001)。结论波形蛋白在胶质瘤患者脑脊液中的水平及脑脊液游离细胞中表达均升高,迁移能力强;脑脊液波形蛋白浓度对预测胶质瘤存在及恶性程度有一定临床价值。
Objective To explore the level of vimentin( VIM) in cerebrospinal fluid of patients with brain glioma and its clinical significance.Methods Enzyme-linked immunosorbent assay( ELISA) was used to detect the VIM level among 50 patients with brain glioma including 22 cases of WHOⅠ-Ⅱgrade and 28 cases of WHOⅢ-Ⅳ grade(glioma group),and 23 patients with brain trauma or cerebral hemorrhage( control group) .Western Blot was used to detect the expression of VIM in free cells of cerebrospinal fluid in the group of WHOⅠ-Ⅱgrade,group of WHOⅢ-Ⅳgrade and control group.Transwell method was adopted to analyze the migration ability of tumor cells in cerebrospinal fluid in the glioma group.Results The levels of VIM in cerebrospinal fluid were in sequence of group of WHO Ⅲ-Ⅳgrade〉group of WHOⅠ-Ⅱgrade〉control group(P〈0.05).Spearmen correlation analysis showed that the level of VIM in cerebrospinal fluid positively correlated with the malignancy grade of glioma(r=0.783,P〈0.001).The expression of VIM in free cells of cerebrospinal fluid was significantly higher in the group of WHOⅠ-Ⅱgrade or WHOⅢ-Ⅳgrade compared with that in the control group,and the increase in the group of WHOⅢ-Ⅳgrade was more significant than that in the group of WHOⅠ-Ⅱgrade.The number of migrated tumor cells in cerebrospinal fluid of group of WHOⅢ-Ⅳgrade was more than that of group WHOⅠ-Ⅱgrade((91.83 ±3.55) vs.(51.37 ±3.35), P〈0.001).Conclusion Both the level of VIM in cerebrospinal fluid and expression of VIM in free cells of cerebrospinal fluid are increased among patients with brain glioma,and VIM has a good ability of migration.The concentration of VIM in cerebrospinal fluid has the clinical value to predict the existence and malignancy grade of brain glioma.
