神经生长因子在人脑胶质瘤及脑脊液中的表达及意义 被引量:2

Expression and significance of nerve growth factor in glioma and cerebrospinal fluid

摘要 目的:研究神经生长因子(nerve growth factor,NGF)在脑星形胶质瘤组织及脑脊液中的表达特点,探讨其与肿瘤病理分级、实体肿瘤部位的关系,以及在肿瘤发生过程中的作用。方法:收集86例星形胶质瘤组织及术前脑脊液标本,取同期10例严重脑外伤行减压术患者的脑组织及10例无神经系统疾病史因其他原因而行腰麻手术患者的脑脊液分别作为对照组脑组织和对照组脑脊液。通过免疫组织化学染色观察NGF在胶质瘤细胞及对照组脑组织细胞中的表达,并采用ELISA法检测NGF在胶质瘤患者及对照组脑组织和脑脊液中的表达水平。结果:胶质瘤组织及脑脊液中NGF表达水平明显高于对照组(P<0.05),高级别胶质瘤中NGF表达水平明显高于低级别胶质瘤(P<0.05)。NGF在胶质瘤组织中主要分布于细胞质及细胞核,在对照组脑组织中主要定位于细胞质。高级别星形胶质瘤中NGF核阳性率明显高于低级别星形胶质瘤,差异有统计学意义(P<0.05)。小脑幕上胶质瘤阳性率明显高于小脑幕下胶质瘤(P<0.05)。结论:NGF在胶质瘤中的表达水平与胶质瘤的发生发展密切相关。 Objective: To study the expression and clinical significance of nerve growth factor(NGF) in glioma and cerebral spinal fluid (CSF). Methods: A total of 86 cases of glioma and CSF were determined for NGF expression by immunohistochemical method (Envision) and ELISA. Ten cases of tissue samples from patients with traumatic brain injury who had received decompression surgery and 10 cases of CSF sam- ples from patients underwent spinal anesthesia without nervous system disease were taken as the correspond- ing control groups. Results: In glioma and CSF cases, the NGF expression levels was higher than that in corresponding two control groups, the difference was statistically significant ( P 〈 0.05 ). There was signifi- cant difference in NGF expression between different grades of glioma ( P 〈 0.05 ) ; the expression of NGF in- creased in parallel with the increase of pathological grade of glioma ( P 〈 0.05 ) ; and there was also signifi- cant difference in glioma incidence between the supratentorial tumour and infratentorial tumour(P 〈 0.05 ). Conclusion: NGF expression was a consistent feature of cerebral tumor, especially in glioma, and its ex- pression was correlated with the genesis and development of glioma.
