摘要
旨在对猪丹毒杆菌的SpaA基因进行PCR检测,并对该基因所编码的氨基酸序列进行生物信息学分析。首先对猪丹毒杆菌SpaA基因进行PCR检测,后将SpaA基因的保守区域进行生物信息学分析。结果表明,PCR扩增得到了与猪丹毒杆菌SpaA基因大小一致的1 531bp片段,二级结构的预测结果显示,该蛋白主要以螺旋、无规则卷曲为主,只有少数的伸展链。推测该蛋白有4个B细胞抗原表位区域。该研究为猪场猪丹毒杆菌病的检测提供了试验数据,也为阐明猪丹毒杆菌病的免疫机理和制备表位疫苗等奠定理论基础。
The aims of this study was to detect the SpaA gene of Erysipelothrix rhusiopathiae, and then analyze the bioinformatics of amino acid sequence encoded by SpaA gene. Firstly,the SpaA gene was amplified by PCR,then the conservative areas of SpaA gene was analyzed by using bioinformatics methods. The results showed that the SpaA gene is 1 531 bp,the predication of secondary structure of the SpaA showed the protein are mainly composed of helix, random coil,and contain a small amount of strand chain. It was supposed the SpaA protein contain 4 B cell antigen epitope regions. The study provided the empirical data for the detection of Erysipelothrix rhusiopathiae at piggery,and at the same time laied theoretical basis to analyze the immunomechanism of Erysipelothrix rhusiopathiae and preparation for epitope-based vaccine.
