摘要
为阐明SpaA在猪丹毒丝菌致病中的作用,以临床分离的猪丹毒丝菌野生强毒株为材料采用DNA重组技术构建spaA基因敲除载体,并利用该敲除载体通过同源重组敲除强毒株的spaA基因,比较强毒株和敲除株对小鼠毒力的差异.结果表明敲除株相比野生株对小鼠的毒力降低102倍,这说明SpaA与猪丹毒丝菌的致病性有关.
In order to elucidate the pathogenic effect of SpaA,the spaA gene knockout vector was constructed from clinical isolated Erysipelothrix rhusiopahiaes wild virulent strain by DNA recombination technology.The spaA gene of the strong virulent strain was deleted by homologous recombination technology and then the virulence difference of strong virulent strains and the knockout strains was compared in mice.The result show that the virulence to mice of knockout strains decreased over 102-fold compared with wild strains.The data indicated that SpaA was related to the pathogenicity of Erysipelothrix rhusiopahiaes.
