摘要
该文建立了HPLC同时测定狭基线纹香茶菜水溶性总黄酮中6种碳苷黄酮含量的方法,并对狭基线纹香茶菜水溶性总黄酮进行抗肿瘤活性评价。采用Kromasil 100-5 C_(18)(4.6 mm×250 mm,5μm)色谱柱;流动相为甲醇-乙腈-0.5%甲酸梯度洗脱;体积流量0.8 mL·min^(-1),检测波长334 nm;柱温为室温(25℃)。以HepG2细胞为供试细胞株,采用MTT法评价水溶性总黄酮的抗肿瘤活性。新西兰牡荆苷2、新西兰牡荆苷3、异夏佛塔苷、夏佛塔苷、牡荆苷和芹菜素-6,8-二-C-α-L-吡喃阿拉伯糖苷线性范围分别为0.25~2.53,0.12~1.20,0.37~3.69,0.16~1.63,0.19~1.92,0.14~1.42μg;平均回收率(n=6)分别为99.6%,100.2%,99.6%,97.9%,98.8%,98.6%,RSD分别为0.87%,2.0%,1.8%,1.5%,1.2%,1.2%。以IC_(50)值作为评价指标,测得狭基线纹香共菜水溶性总黄酮在给药24,48,72 h后抑制HepG2细胞增殖的IC_(50)值分别为1.89,1.71,1.51g·L^(-1)。本法快速、简便、准确,重复性好,可用于狭基线纹香共菜水溶性总黄酮的质量控制。狭基线纹香茶菜水溶性总黄酮对HepG2细胞增殖有一定的抑制作用。
This research established an HPLC method for determination of six C-Glycoside flavones of warer-soluble total flavonoids from Isodon lophanthoides vat.gerardianus(Benth.)H.Hara,and studied the antitumor activity of the warer-soluble total flavonoids.The HPLC system consisted of Kromasil 100-5 C_(18)(4.6 mm×250 mm,5μm)column and a solution system of methanol,aeetonitrile and 0.5%formic acid gradient elution at a flow rate of 0.8 mL·min-1 and the wavelength of detector was at 334 nm.The column temperature was 25℃.The antitumor activity of water-soluble flavonoids was assayed using HepG2 cell as the tested cell.The linear ranges of viceninⅡ,viceninⅢ,isoschaftoside,sehaftoside,vitexin,6,8-di-C-α-L-arabinosylapigenin were 0.25-2.53,0.12-1.20,0.37-3.69,0.16-1.63,0.19-1.92,0.14-1.42μg,respectively.The average recoveries(n=6)were 99.6%(RSD 0.87%),100.2%(RSD 2.0%),99.6%(RSD 1.8%),97.9%(RSD 1.5%),98.8%(RSD 1.2%),98.6%(RSD 1.2%),respective-ly.After exposure in 24,48,72 h,the total flavonoids showed inhibitory effect on the proliferation of HepG2 ceils with IC50 as the evaluation index,the IC50 values of 1.89,1.71,1.51 g·L-1,respectively.The method is quick,simple and accurate with good're-producibility,and can be used for determination of viceninⅡ,viceninⅢ,isoschaftoside,schaftoside,vitexin,6,8-di-C-α-L-arabino-sylapigenin in the warer-soluble total flavonoids from I.Iophanthoides var.gerardianus.The warer-soluble total flavonoids from I.loph-anthoides have inhibitory effect on the proliferation of HepG2 cells.
