摘要
目的通过检测肿瘤坏死因子受体相关因子(TRAF-2)在骨肉瘤细胞表达的变化及三氧化二砷(As2O3)和血管内皮细胞生长因子(VEGF)对其表达的影响,探讨TRAF-2在骨肉瘤发病机制中的作用。方法人骨肉瘤Sa OS-2细胞经分别加入不同浓度的As2O3和(或)VEGF培养48 h后,采用实时荧光定量PCR法测定TRAF-2 mRNA的表达水平。结果 100 ng/ml浓度的VEGF能显著上调TRAF-2 mRNA的表达水平,差异有统计学意义(P<0.01),50 ng/ml以下浓度的VEGF对TRAF-2 mRNA的表达水平没有影响,差异无统计学意义(P>0.05);2μmol/L以上浓度的As2O3能显著下调TRAF-2 mRNA的表达水平,并能显著降低100 ng/ml浓度的VEGF对TRAF-2 mRNA表达水平的上调功能,差异有统计学意义(P<0.01);而1μmol/L浓度的As2O3对TRAF-2 mRNA的表达水平没有影响,并不影响100 ng/ml浓度的VEGF对TRAF-2 mRNA表达水平的诱导作用,差异无统计学意义(P>0.05)。结论 TRAF-2在骨肉瘤的发病机制中可能起重要作用,VEGF具有上调TRAF-2 mRNA表达水平的作用,而As2O3则起相反作用,两者具有一定的量-效关系。
Objective To explore the possible roles of tumor necrosis factor receptor-associated factor-2( TRAF-2) in the pathogenesis of osteosarcoma,and the effects of arsenic trioxide( As2O3)or vascular endothelial growth factor(VEGF) on human Sa OS-2 cell line were observed. Methods Human osteosarcoma Sa OS-2 cell line was cultured and treated with As2O3and(or) VEGF at different concentration. After 48 hours,real-time quantitative reverse transcription polymerase chain reaction(QRT-PCR) methods were used to investigate the level of TRAF-2 mRNA expression. Results The expression of TRAF-2 mRNA was significantly increased in Sa OS-2 cell line treated with VEGF at 100 ng / ml concentration,the difference had statistical significance( P〈0. 01),while no change of TRAF-2 mRNA expression was observed under 50 ng / ml concentration of VEGF( P〉0. 05). Moreover,it was significantly dicreased of TRAF-2 mRNA expression in Sa OS-2 cell line treated with As2O3 at 2 μmol / L concentration and more( P〈0. 01). Interesting,the inducing function of VEGF at 100 ng / ml concentration on the TRAF-2 mRNA expression can be inhibited significantly by As2O3 at this concentration( P〈0. 01). Whereas,no change of TRAF-2 mRNA expression was seen when treated whith As2O3 at 1 μmol / L concentration( P〉0. 05). In addition,As2O3 at 1 μmol / L concentration couldn't also affect the TRAF-2 mRNA expression in Sa OS-2 cell line treated with VEGF at 100 ng / ml concentration( P〉0. 05),the difference had no statistical significance. Conclusion TRAF-2 may paly an important role in the pathogenesis of osteosarcoma. The osteosarcoma cell proliferation can be promoted by VEGF and inhibited by As2O3 in a dose-dependent manner.
